Mr. Bockosh is an electrical engineer with the U.S. Bureau of Mines In Pittsburgh, and has devoted the last two years to research on underground illumination. He formerly was a mine maintenance foreman and worked two years underground. He is a graduate of the University of Pittsburgh, where he is now working toward his masters degree in mining. When Congress first legislated light in American coal mines, it was a little like mandating a walk on the moon before the first satellite had been put in orbit. It was never a question, of course, of whether it could be done. But no one knew quite how. It's one thing to string a few light bulbs, and quite something else to provide uniform, sufficient, safe and reliable illumination, with fixtures that will hold up under some of the toughest conditions in the world - in the life-and-death circumstances of a craggy, black cavern filled with dangerous, moving machinery. In fact, in 1969 when Congress originally mandated illumination standards for the work places of underground coal mines, no one even knew just how much light was needed. That question was answered within a year by the National Bureau of Standards. In a study commissioned by the U.S. Bureau of Mines, the Bureau of Standards determined that underground mine should be illuminated with a light intensity of 0.06 foot-lamberts, a measurement of light reflected from surfaces in the lighted area-as opposed to foot-candles, a measurement of light from the source of illumination-in order for miners to adequately see what they are doing. That study was followed by more detailed research done at the Bureau of Mines' direction by the Research and Development Department of the Naval Ammunition Depot in Crane, Ind. This work evaluated the specific lighting needs of miners engaged In more than 100 different job tasks in all three types of coal mining- conventional, continuous and longwall. The Naval Depot report brought forth nearly 300 pages of detailed lighting data and concluded that proper illumination of mine work areas would serve more than just the cause of mine safety.

"The Bureau of Mines receives and answers many requests for information on the qualities and properties of American coals, and also publishes many papers that deal with coal characteristics. Compilations of chemical analyses of coal from specific locations are contained in a series of technical papers,2/ by States, whereas typical analyses of coal seams, by States and counties, are given in data books, volumes 1 to 7,2/ by producing districts, as defined in the Bituminous Coal Act of 1937. Bulletin 446, ""Typical Analyses of Coals of the United States,"" condenses and supplements these data books, which do not apply to the Western States. Other coal properties are described in numerous publications listed in the Bureau of Mines Bibliography of Publications. However, no recent publication lists average B.t.u. values by rank on a Statewide and national scale. Averages have been estimated in the past, and some of them are included in this paper for comparison.The average and the ranges of as-received B.t.u. values of American coals on a Nation-wide basis, as found by the statistical method described in this paper, are given in table 2 and on a State-wide basis in table 3. The following is abstracted from table 2:In deriving average B.t.u. values for coals, the results depend on the type of data available and the methods of calculation. The data, or analyses, may represent any of three types of samples - mine, tipple, or delivered. In mine sampling, a channel cut is made of a vertical face of the coal bed, carefully excluding most of the impurities, and the material retained is the sample. It is readily seen that mine samples are selected samples and do not strictly represent the unmined coal bed, unless the coal contains no major impurities. Mine samples generally represent the best-quality coal that could be produced. In contrast to mine samples, tipple and delivered samples represent coal that actually has been mined, and their analyses may show either lower or higher values than mine samples of the same coal, depending on treatment and preparation after mining.Of the thousands of analyses accumulated by the Bureau of Mines, most are of either tipple or delivered samples and represent the coal after it has been mined. Any average calculated from this aggregate of data will, consequently, reflect coal that has been produced, and not unmined coal in situ.The method of calculating an average from the basic data also will have considerable influence. If there are three seams of coal in a county, the average analysis for the county is not necessarily the arithmetic mean of analyses of the three individual seams. For a probable average of unmined coal, the B.t.u. values of each seam should be weighted in proportion to the unmined reserves of each seam. In similar manner, an average for past pro¬duction should be weighed according to the past production for each seam. Figures for coal reserves are only estimates based upon geologic evidence and its interpretation. Production records are much more accurate than estimated reserves, although detailed production records for individual seams by counties are very few."

This surveillance report summarizes data on work-related fatal and nonfatal injuries, illnesses, and hazardous exposures in the mining industry for the 10-year period 1986-1995. The term "surveillance," as used in public health, may be new to many readers of this report. With regard to occupational safety and health, the goal of surveillance is to describe the occurrence of work-related injuries, illnesses, and known hazardous exposures; to identify new hazards that may occur due to the introduction of new technology or other factors; and to assess the effects of preventive measures designed to improve worker safety and health. Surveillance provides an overall picture, which can be used to focus resources on areas most in need of prevention programs or further research. Surveillance generally requires the use of multiple sources of data, all of which have their own strengths and limitations. Surveillance in the U. S. mining industry is complicated by the fact that U.S. national data systems that provide information on mining are based on two different definitions of the mining industry. One is established by the Mine Safety and Health Administration (MSHA), the other by the Standard Industrial Classification (SIC) System [Office of Management and Budget 1987] and the 1980 U. S. Bureau of the Census (BOC) Classification System [U.S. Bureau of the Census 1982]. The mining industry as defined by MSHA is based on the regulatory jurisdiction of that agency. Reporting requirements for injuries, illnesses, and workplace exposures are stipulated under the Federal Coal Mine Health and Safety Act of 1969 and the Federal Mine Safety and Health Amendments Act of 1977. Of note is that the oil and gas extraction industry falls outside of MSHA jurisdiction. The Occupational Safety and Health Administration (OSHA) was charged with enforcement in the oil and gas industry under the Occupational Safety and Health Act of 1970. Under the SIC, the definition of mining covers the extraction of naturally occurring minerals, including liquids (petroleum), gases (natural gas) and solids (coal, metal, and nonmetallic minerals). Exploration and development of mineral properties are also included in the SIC definition of the mining industry. The SIC was developed by the Office of Management and Budget for use in classifying establishments based on the type of economic activity in which they are engaged and serves as the industry classification standard for all establishment based Federal economic statistics. This classification is important for occupational safety and health surveillance because it also serves as the industry classification system for all data on work-related injuries and illnesses collected by the U.S. Bureau of Labor Statistics (BLS). The U.S. Bureau of the Census uses an industry coding scheme similar to that used by the SIC. For the mining industry, the categorization is identical in both systems. Death certificate data made available by the National Center for Health Statistics (NCHS) are categorized under this system. Death certificate data are particularly important for surveillance of mortality from work-related illnesses. Key differences between the SIC/BOC categorization system and the MSHA system can be summarized as follows: • MSHA excludes the oil and gas industry, which is classified as part of the mining industry under SIC/BOC. • MSHA excludes work that is done off of mine property; work off of mine property is included under SIC/BOC if it is performed by an establishment falling into one of the SIC/BOC mining industry codes. • MSHA includes mine-associated mills and processing plants; these are classified under the manufacturing industry, rather than mining, according to the SIC/BOC coding structure. Since the data collected by MSHA and the data collected by BLS and NCHS using the SIC/BOC classification system are both important sources of information on the mining industry and in many cases serve different purposes, data from both systems are presented in this report. In general, data using the SIC/BOC system are comparable across U.S. industries and should be used when comparing mining to other U.S. industries. Much of the data in this report are presented in the form of frequencies or rates. A frequency provides only a count of the number of cases without providing any information about the risk of occurrence. Rates provide an estimate of the risk of injury or illness. The reader is cautioned, however, that rates based on small numbers can be unstable; thus, any inferences should be drawn with care. In this report, no rates are presented for groups in which there were fewer than three cases. The incidence of most injury and illness conditions differs by age, sex, and race. For this reason, data in many surveillance reports are adjusted for these demographic factors. It was not possible to do this for most of the rates in this report because information on demographics was not available. With the exception of the proportionate mortality ratios (PMRs), all data presented are unadjusted. A detailed account of the sources of data is in appendix A. The methods used in data selection and analysis are described in appendix B.

The first steps in the process of mine evacuation are the recognition of a problem and an attempt to communicate the problem to miners who may be affected. This chapter will focus on the way that a problem, fire, came to the attention of mine personnel and the messages that were sent to miners in the affected areas. The concept of information uncertainty, which was introduced in chapter 4, will be discussed as it influences problem perception and diagnosis. Sociotechnical and interpersonal communications will be explored and suggestions for improving these systems will be offered. It might seem that the first indication of a dangerous fire would motivate individuals to take self-protective action, to evacuate the affected area or structure, and to provide clear warning to others who are in danger. Research has actually shown that in most situations this does not occur. Instead, time is taken to gather more information, confirm information that is provided, and consider possible alternative explanations that could account for the given circumstances [Canter 1990; Scanlon 1979; Bickman et al. 1977]. This process of confirmation can lead to the loss of critical time. Canter [1990] summarizes the problem in his book, which reports studies of a number of fire events: "As discussed throughout this book, ambiguity and confusion, incoherent instructions and time-wasting actions, lack of appropriate instructions and misunderstanding of the nature of the event that is unfolding, are all hallmarks of fires and emergencies that kill people." In this chapter, the detection of each mine fire and the communication of warning to endangered miners will be reviewed with an emphasis on how those processes were affected by the availability and use of information. Information can become available through a variety of mechanisms during an emergency situation. First, cues may be taken directly from the environment. Smoke is an obvious example. Secondly, mechanical devices, such as smoke detectors, may provide warning messages. A third source of information is interpersonal communications. These can occur face-to-face or through some mechanical device such as a telephone. With all three methods there is a possibility of miscommunication, misunderstanding, and misinterpretation. All of these means of communication were used with varying degrees of success in the three mine fires analyzed here. A fire, like any nonroutine situation, engenders uncertainty about a diagnosis and understanding of the problem [Mead 1938]. This uncertainty leads to delays in realization of the seriousness of the situation and therefore in the proper response to it. Delay in action is an important concern in any fire setting, but is even more at issue in underground coal mine fires. Mine fires are qualitatively different from structural blazes: workers' escapeways may be miles long; the seam height at many operations is so low that it is impossible to walk upright; access to underground workings is always limited to a few (sometimes only two)

Underground coal winning operations can broadly be classified as conventional, continuous, and longwall mining. In conventional mining, the operations of cutting, drilling, blasting, loading and hauling, and roof support are performed in that sequence in each working face in a work area. The major difference between continuous and conventional mining is that the coal is cut and loaded by a continuous miner in the former, thereby combining the unit operations of cutting, drilling, shooting, and loading into one. Longwall applications have been viewed with great interest in recent years, and although at the end of 1970 there were only 37 longwall installations in the U. S. with combined production of less than 4 percent of underground total, there is no question about the growth potential of longwall in the United States. Increased capacity, versatility, and power characterize continuous miners today. Although continuous miners have outsold conventional gathering-arm type loading machines for some years, the latter, in the new extra-high capacity types, are continuing to find wide use. Whether the mining system is continuous or conventional, the haulage units may include six-wheel articulated shuttle cars, tractor-trailer units, or extensible belt and bridge-conveyor systems, all tending to increase the productivity of the face units. The decision as to which to choose - continuous or conventional mining - is complicated, and both systems are capable of high production rates. "It is evident that the incidence of coal worker's pneumoconiosis (CWP) or black lung disease varies in different coal producing regions of the United States. Many people have hypothesized about the reasons for the differing occurrences. Because underground coal mining involves many mining methods under changing geological conditions, several factors may be involved in contracting CWP such as: the amount of quartz in the respirable dust, the trace elements in the respirable dust, the rank of the coal that produced the respirable dust (bituminous, anthracite), the amount and size of respirable dust, etc.Recognizing the uniqueness of underground coal mining and CWP, research investigations involving seam characteristics, mining system and worker position are being conducted in the center to establish standard procedures for characterizing some coal properties that may be involved in workers contracting CWP. This has been accomplished by the in-mine sampling of airborne dust at various worker locations and mining system configuration and by performing laboratory analyses of mined material (coal and rock) taken from the mines. (FIGURE 49 and 50)The investigations includes a method of classifying coal seams according to their potential to generate respirable dust. It includes statistical analyses of the size and locational variations of the elemental compositions of respirable coal mine dust in operating underground mines from seams located in the eastern, midwestern, and western United States, and is currently completing research which is investigating the relationship between the elemental compositions of respirable dust sampled near a continuous-mining machine and a laboratory-produced respirable dust."

[The meaning of the term "bauxite' has always been somewhat vague and indefinite, and the term has been used in various applications to denote sometimes a rock, sometimes a mineral, but more frequently an ore. It seems logical to think of bauxite as an ore, however, because a very large part (probably at least 85 per cent) of the entire world's production of bauxite is at present being used as a source of aluminum and aluminum chemicals. In the United States there are two districts that, produce bauxite - the Arkansas district and the Alabama-Georgia-Tennessee-Mississippi district. The deposits in Mississippi have been discovered recently and are riot as yet productive but it seems reasonable to consider them as apart of the-district hat includes the deposits of Alabama, Georgia, and Tennessee. The, two districts are often referred to as the Arkansas field and the Appalachian field. Of these two the Arkansas field has been by far the largest producer, and at present over 90 per cent of the bauxite production of they United States comes from the Arkansas field. The bauxite industry in the Appalachian field had its beginning in 1889, and since that time the total production of the states of Alabama, Georgia, and Tennessee has been, nearly a million tons. The production of bauxite in this field has languished in the last five, or six, years, and In 1927 its production was only about 14,000 tons, according to Harder4. The recent decrease in production is due mainly to the fact that many .of the known deposits of commercial-grade ore have become exhausted. There are, however, considerable quantities of low-grade bauxite in the Appalachian field, but these have not been available because there has been no satisfactory way of concentrating low-grade bauxite. The quantity, of low-grade material in the Appalachian field is probably, fully as large as the tonnage of commercial ore which the field has already produced.]

"The town of Ajo is built in the desert and consisted of but a few houses when the New Cornelia Copper Co. started operations. Since there were no stores at Ajo or vicinity the New Cornelia Cooperative Mercantile Co. was established to serve the needs of the employees of the New Cornelia and allied companies.The New Cornelia Copper Co. advanced the money to erect the store building and to buy the stock, and assumes all risks. In all, $85,000 has been advanced, representing the capital stock of the enterprise.The store is run by a store manager appointed by the Copper Company and who reports only to the general superintendent. A committee of seven workmen, representing the various depart¬ments, meet with the store manager monthly and make suggestions for the betterment of the service and, present complaints which have been brought to their attention.The store is well managed, carries a large and well assorted stock and is modern in every respect. Prices are at the same level as in the other Southwestern mining camps. The Copper Company does not particpate in the profits of the store but does receive six per cent interest on the capital advanced. At the end of the year, after making proper deductions for interest on capital account, depreciation of store equipment and fixtures, and, for a proper sum to the surplus account, the rest of the profits is distributed among the employees of the company in proportion to the amount of their purchases."

The International Organization for Standardization (ISO) standard ISO 2631 [1,2] provides three methods for evaluation of human exposure to vibrations that contain occasional shocks or impacts. Part 1 of the standard specifies the running r.m.s. or maximum transient vibration method (MTVV) and the fourth power vibration dose value (VDV). Part 5 of the standard provides a method of computing the stress in the lumbar spine for humans exposed to multiple shocks. Alem et al [3] have reported application of these methods to data for tactical ground vehicles. This paper reports and compares VDV and spinal stress evaluations of more than 90 hours of vibration and shock measurements on North American locomotives engaged in through freight operations. The measurements evaluated in this paper were obtained for full crew shifts on 19 freight locomotive runs on mainline track in locations from New York to California. The shifts ranged in duration from 187 minutes to 497 minutes. The average speeds for the shifts were from 21.0 mph to 54.6 mph. All measurements were made on locomotives hauling freight trains in regular revenue service. Data Acquisition and Processing The results reported here were computed using test data acquired from a tri-axial seat pad, accelerometer at a sample rate of 400Hz with an anti-aliasing filter corner frequency of 100Hz. The VDVs and the lateral and longitudinal spinal stress values were computed directly from the acquired test data according to the procedures specified and described in [1] and [2]. The vertical spinal stress values were computed by converting the as-acquired test data to a sample rate of 160Hz for input to the vertical spine model, as required in [2]. The conversion of the test data from the as-acquired sample rate of 400Hz to the required sample rate of 160Hz involved up sampling or interpolating the test data to an equivalent sample rate of 800Hz, band limiting the resultant data with a low-pass filter corner frequency of 60Hz and finally down sampling or decimating the 800Hz data to a sample frequency of 160Hz. Discussion The vertical VDVs computed according to [2] for the 19 shifts ranged from 2.68 to 9.33 m/s1.75. In all but one case, the vertical values were greater than the values for the lateral or longitudinal directions. Note that the health guidance in [1] puts the lower boundary of the health guidance caution zone at a VDV value of 8.5 and the upper boundary at 17 m/s1.75. The daily equivalent static compression dose computed following [2] ranged from 0.123 to 0.434 MPa. Health guidance provided in [2] states that there is a low probability of an adverse health effect if the daily dose is less than 0.5 MPa.

Develop quieted, hand-held pneumatic percussion drills that can replace standard hand-held drills in many underground mining applications. The Problem Percussion drills are the noisiest machines used in underground mines; noise levels of 115 to 120 dBA are common, requiring the use of hearing protectors and limiting the time a miner is permit-ted to operate the drill during an 8-hour shift. Hand-held pneumatic percussion drills pose a particularly difficult problem because the operator cannot be isolated from the noise source (e.g., an acoustical cab), and alternative technologies for drilling roof bolt holes and blastholes in tight quarters are not readily available.

INTRODUCTION The National Institute for Occupational Safety and Health (NIOSH) at its Pittsburgh Research Laboratory (PRC) is interested in the application of water mist sprays to extinguish diesel fie1 fires in underground mine diesel refueling areas. To this end experimental studies have been carried out on the extinguishment of diesel fuel fires at PRL's large enclosed fire suppression facility (FSP) at its Lake Lynn Laboratory (LLL). Two sizes of shallow fuel pans (3x3 ft and 5x7 ft) contained the fires, and a number of commercial mist spray nozzles (at roof locations) and spray/fire locations were tested (52 fires in all). Visual observations of the pool fire extinguishment process by the overhead sprays with an infra-red sensitive videocom suggests that there are two distinct actions involved in extinguishment due to two different transport mechanisms by which the water droplets can enter the fire plume. First, is a direct injection of water droplets into the plume from overhead. This is believed to involve larger water drops and leads to an extinguishment time of several minutes. Second, is an indirect injection process involving small water drops which are entrained in the sideways air flow that feeds oxygen to the fire. This second process apparently leads to relatively fast quenching of the fire in less than 1 minute - a highly useful attribute for a fire protection system. This paper describes a relatively simple parametric model for the indirect water injection process, and defines the critical spray conditions for achieving rapid quenching of the pool fire. While the model is consistent with the pool fire data obtained to date, the PRL studies are continuing with more complex pool fire scenarios. It remains to be seen if this simple parametric approach applies to these future fire tests. THEORETICAL Critical Drop Size In the two-dimensional representation of figure I, an oil pool fire is considered to be a cone whose base is the vaporizing liquid oil surface having an area n?, above which exists a fire plume diffusion flame) of height, L. The rising plume induces a horizontal air flow into the side of the plume supplying the oxygen necessary for propagating gas phase combustion reactions. Water droplets suspended in the air around the plume will tend to follow the horizontal air flows into the plume. "Part IV.- The Resistance Caused by Right-Angle Bends. IntroductionThis paper deals with the fourth phase of an extensive research on coal-mine ventilation factors. A comprehensive report of the entire work is in course of preparation, but abstracts of the various chapters are being prepared in order to make the major results public as rapidly as possible. Parts I, II and III were issued as Serials Nos. 2621 (July 1924), 2647 (October 1904) and 2671 (March 1925) and dealt with the resistance of unobstructed entries, the resistance due to mine cars and, the resistance of timbered entries respectively.Description of the Tests With the last cutthrough between the two butt entries of the test section in the Experimental Mine open, the air flow was deflected, around two right-angle bends in the same direction, the butt entries being 50 feet center to center and approximately 6 by 9 feet in cross section. Each bend had a dead end opening off of it; that at the first bend was in the direction of flow and that at the second bond was at right angles to the flow. For a primary test condition the inner corner of each bend was trimmed square and the dead ends were closed off with board brattices set flush with the line of the cut-through.The two corners of the first band were, then modified both independently and in conjunction. Various modifications of both bends in conjunction were also tested with rather surprising results as to the effect of conditions at the first bend, upon the pressure losses at the second. To confirm the theory developed to fit these results a series of tests were made to give a direct comparison of the -pressure losses as between two bends with the air flow deflected in the some direction at each and with the air flow deflected in opposite directions. The former is designated ""double"" and the latter a ""reversed"" right-angle bend. This series of tests was conducted, however, on the bends at one of the intermediate cutthroughs at which the flow could be deflected us either direction at the second, bend. Here also the average cross-section was 6 by 9 feet and the bends were 50 feet apart."

Native asphalt was produced in 1980 by six companies in four States. Leading States were Texas and Utah. Output decreased 22% in 1980 to 1.25 million tons while value decreased 2% to $25.0 million. Bituminous limestone was produced by Whites Uvalde Mines and by Uvalde Rock Asphalt Co. in Uvalde County, Tex.; by Southern Stone Co. in Colbert, Ala.; and by Barton County Rock Asphalt Co. in Barton County, Mo. The product was used mainly in street and road repair. Gilsonite was produced by American Gilsonite Co. in Uinta County, Utah, and by Ziegler Chemical and Mineral Corp. in Weber County, Utah. This material was used for purposes other than road repair.

"Incidental to a cooperative investigation with the Carnegie Institute of Technology and the International Combustion Engineering Corporation on the burning characteristics of a powdered coal and the powdered semi-coke therefrom, the U. S. Bureau of :alas has determined the relative temperatures at which rapid self-heating occurs in these two fuels. Determinations were made on various screen sizes of the two fuels. Chemical analyses on different sizes were made in the bureau's Coal Analysis Laboratory, of which H. M. Cooper is in charge. The true densities were determined b the method given in Bureau of Mines Technical Paper 8.4The apparatus used in the determination of the temperatures for rapid self-heating was similar to that used by Wheeler A 10-gram sample of the fuel was placed upon a screen in a glass tube enclosed in a vertical electric furnace. The tube was surrounded with sand. One thermocouple -as imbedded in the sand and another in the fuel. Measurements of the temperatures were obtained at regular intervals with a recording potentiometer, as the furnace temperature was uniformly raised. Dry air was passed down through the fuel samples at a uniform rate of 123 c. c. per minute; the temperature of the air was substantially that of the furnace."

Although early Spanish settlers used coal several centuries ago, significant commercial coal mining in New Mexico began in 1861, when the U.S. Army opened a mine to supply Fort Craig. Since then, coal from the state has been used to fuel railroads, lead and copper smelters, and other local industry. In recent years, lower-cost strip mining methods and an increased demand for coal by electric utilities in the Southwest have caused an increase in New Mexico coal production. The coal-bearing regions of New Mexico underlie about 25,000 square miles or20.6 percent of the total area of the state. However, the majority of coal-bearing regions lie under Indian lands that are regulated by the Office of Surface Mining (see Navajo and Hopi data on pages 72-75). Today, most of the coal produced in New Mexico comes from the San Juan Basin in the northwest part of the state and the Raton Area in the north central part. New Mexico's reserves are estimated to be 4.6 billion tons, about one percent of the national reserve. New Mexico's permanent regulatory program was conditionally approved December 3 1, 1980. Its Abandoned Mine Lands Program was approved July 17,198 1.

"In connection with the proposed use of Cordeau-Bickford to fire L.O.X. (liquid oxygen explosives) in well drill holes, the question has arisen as to the effect of very low temperatures on (1) the rate of detonation of Cordeau-Bickford; (2) the sensitiveness to detonation of its TNT filling; and (3) the lead covering of Cordeau-Bickford.The Cordeau-Bickford used in this investigation was the standard 6 mm. size, such as is regularly used in blasting, and was manufactured and furnished by the Ensign-Bickford Company, Simsbury, Conn. All pieces tested were taken from rolls, all of which were from the same melted batch of TNT in order that all the pieces tested might be as nearly identical as possible.Different portions of this Cordeau-Bickford were tested plain (not countered), countered, and double countered. The countering and double countering was carried out by the manufacturer prior to shipment.The determination of the temperature at the instant of making a test was accomplished by the use of a Leeds and Northrup potentiometer and copper constantan thermocouples. Each couple was made from No. 28 B & S gage wire with short junctions. One junction was inserted in a hole drilled by a No. 60 drill through one lead wall of the Cordeau-Bickford to approximately the opposite wall, so that this junction was in the center of the TNT filling while the other junction was immersed in ice water. The potentiometer readings in millivolts were converted to °C. from the calibration curve of the thermocouple wire,"

Pulmonary inflammation is accompanied by increased microvascular permeability and leakage of plasma into alveolar spaces. Serum has been shown to affect superoxide (O2-) production from pulmonary alveolar macrophages (PAM) in vitro, and in some cases serum has been shown to be necessary for stimulation of PAM. An electro-optical method was used to measure O2- production from single PAM by measuring the reduction of nitroblue tetrazolium (NBT) to a diformazan precipitate (NBTH2) from videorecorded images of individual cells. PAM were stimulated by their initial attachment to culture dishes independent of serum. The addition of 2% serum to the culture medium increased NBTH2 production during attachment, whereas concentrations of 5 and 10% serum had no additional effect. In contrast, PAM incubated for 2 hrs could not be stimulated by phorbol myristate acetate (PMA) or zymosan particles to produce O2- unless serum was present during the incubation and/or stimulation; however, NBTH production was similar in all cases of serum addition. Also2, serum alone did not stimulate adherent PAM indicating that serum is necessary but not sufficient for stimulation of attached. PAM. These results suggest that changes in microcirculatory permeability may have a pronounced effect on superoxide production by PAM and that serum may help regulate or condition these cells during inflammation of the lung. The mucus secreted in the respiratory tract provides the first barrier against inhaled particulate and gaseous toxicants. Trachea removed from pathogen-free rats were maintained as organ cultures and used to study the effects of coal dust exposure on the synthesis of mucin. The high molecular weight isotopically labeled (3H-glucosamine, 14Cleucine or 35S-sulfate) mucin could be purified by gel filtration, treatment with testicular hyaluronidase, ion exchange chromatography, delipidation and CsBr density gradient centrifugation. To examine effect of coal dust on mucin production, groups of explant cultures were exposed to media containing coal dust at 100 µg per ml every 2 days for 2 weeks while control cultures were treated with media without dust. Analysis of the spent culture media showed that treatment with dust markedly decreased the production of nondialysable glycoproteins as well as hyaluronidase-resistant acid-precipitable fraction consisting mainly of mucin. Since the synthesis of protein was not affected to the same extent the decrease in mucin production is not entirely due to cell death. In separate experiments rats were subjected to in vivo coal dust exposure in inhalation chambers and tracheae of these and control rats were removed for explant cultures. The incorporation of precursor isotopes into mucin by these explant cultures are being examined.
