Occupational exposure to hand-arm vibration through the use of powered hand tools can result in reductions in tactile sensitivity, grip strength and manual dexterity. In fact, even acute exposures to vibration cause shifts in vibrotactile thresholds in exposed fingers (2,4,5). Although reductions in tactile sensitivity after acute vibration exposures are transient, cellular changes associated with this shift in sensitivity could lead to the more permanent reductions in tactile sensitivity that are a common symptom of hand-arm vibration syndrome (HAVS). Methods Animals. Male Sprague Dawley rats (6 weeks of age) were used for all experiments. Animals were housed in AAALAC accredited facilities, and all procedures were approved by the NIOSH Animal Care and Use Committee and were in compliance with the CDC guideline for care and use of laboratory animals. Vibration exposures were performed by restraining rats in a Broome-style restrainer, and securing their tails to the vibration platform using 6 mm wide straps that were placed over the tail every 3 cm. Restraint control animals were treated in an identical manner except that the tail platform was set on isolation blocks instead of a shaker. Tail temperature and current perception thresholds (CPTs). Rats were exposed to 4 h of tail vibration (125 Hz, 49 m/s2), or restraint. Tail temperature was collected prior to and immediately following the exposure using an infra-red camera. Sensory neuron function was assessed by measuring CPTs with a Neurometer (Neurotron, Baltimore, MD). Transcutaneous nerve stimulation was applied to the C10 region of the tail. Three frequencies were used to test specific fiber types (5 Hz – C, 250 Hz - Ad, and 2000 Hz – Aß). The intensity of the stimulus was automatically increased in small increments until the rat flicked its tail. Tests at each frequency were repeated until the animals displayed 2 responses that were within 2 CPT (or 0.02 mA) of each other (2-4 tests/animal). CPT tests were performed prior to the exposure, immediately following the exposure, and 24 h after the exposure. Data Analyses. Temperatures were analyzed using a one-way ANOVA with animal as a random variable. ANCOVAS were used to analyze the CPTs at each frequency. Temperature at the time of the CPT test was used as a covariate, and animal was used as a random variable Results Tail temperatures declined between pre and post exposure (F(1,25) = 62.85, p < 0.001; mean ± sem pre 25.96°C ± 0.42, post 20.71°C ± 0.77), but were back to baseline levels by 24 h after the exposure in both groups of rats. The ANCOVAs demonstrated that exposure to a single bout of vibration did not alter sensitivity of C (5Hz) or Ad (250 Hz) fibers to stimulation.

"Introduction.The theory of fractional ""education"" of oil from oil shale has been used by several inventors as the basis for the design of retorts for the production of oil from the oil shales of the United States. The term fractional ""education"" gives expression to the rather common belief that when an oil shales is heated it produces light, low-boiling oils as low temperatures, and increasingly heavier, higher-boiling oils as the temperature of retort¬ing rises. The theory evidently had its origin in the fact that crude petroleum may be fractionally distilled, yielding light oils at low distillation temperatures, and heavier oils as the distillation temperature increases is as, gasoline is produced from petroleum at relatively low temperatures, kerosene at higher temperature, and heavier products such as gas-oil and lubricating distillation at still higher temperatures.The writers have examined several oil-shale retorts designed to take advantage of the belief that crude, and in some cases refined, gasoline can be recovered from the shale at low temperatures; kerosene at higher temperat.res, and so on. Such retorts are usually designed to operate continuously and have several vapor-pipes lending from them at differing intervals and passing to separate condensers. Designers of such retorts repeat that the distillate from the low-temperature parts of the retort will be gasoline, and that as the shale passes to zones of increasingly higher temperatures, the vapor lines lending from these zones rill remove increasingly heavier products such as kerosene, gas-oil etc."

As longwall shields are lowered in preparation for advance, dust from the canopy falls directly into the air stream with the potential of becoming entrained. Historical dust sampling data from the early 1980s shows that shield advance contributed an average of 12 percent to the airborne respirable dust levels found on longwalls in the United States. Data from the 1990s indicates that shield dust liberation had increased to an average of 23 percent. Increased longwall production levels over this time resulted in the need to move a larger number of shields during each shift, while an increase in air velocities on the face was also occurring. These factors combine to suggest that greater quantities of shield dust are being entrained into the face air. A series of controlled laboratory tests were conducted in a test stand simulating shield dust entrainment to evaluate changes in airborne respirable dust levels at different air velocities. Significant differences in respirable dust entrainment were observed. The test facility, test methodology, and dust sampling results will be reviewed and discussed

"INTRODUCTION ""Free silica"" includes all free forms of silicon dioxide, such as quartz, chalcedony, tridymite, cristbbalite, opal, diatomaceous earth, and lechatelierite. The term, has been used rather loosely however, as synonymous with quartz, as quartz is the most abundant form of free silica. ""Combined silica,"" on the other hand, has been reserved to denote silica combined with other elements as silicates.Improvements in methods for the determination of free silica, and quartz in particular, have been the subject of considerable interest during the past decade, owing mainly to the accented and increased knowledge concerning quartz as a causative factor in silicosis. Analysis for quartz, however, is of importance in many other fields, including the determination of quartz in ceramic raw materials, such as feldspar and clay; the evaluation of a beneficiation procedure by the ore dresser, such as in the concentration of iron ore; control studies of a manufactured product, such as silica brick; and routine analysis of rock, mineral, and ore samples by chemists and petrographers."

A study was made by the Bureau of Mines to determine if shapes of oxide-glass compositions could be roll-formed at high temperatures, and to find the conditions required for adequate formability. A unit with directly connect&apos; furnaces and heated rolls that allowed forming of most of the compositions developed. Glasses of the feldspar type were chemically unreactive with the alumina grains, and allowed the best roll forming. Optimum forming was attained at temperatures of 1,400° to 1,530° C, with compositions containing 30 or 40 percent feldspar and 60 or 70 percent alumina. Formability was better when compositions contained minus 325-mesh materials, rather than coarser or finer powders. In many cases, roll-formed shapes had physical properties equal to or better than the normally sintered shapes of the same composition.

Exposure to whole-body vibration (WBV) and the postural requirements of the job have been identified as important risk factors in the development of musculoskeletal disorders of the back among workers exposed to a vibratory environment. This paper focuses on preliminary results of WBV data collected for two groups of haulage trucks ? four older trucks from manufacturer A (MFR-A) and two newer trucks from manufacturer B (MFR-B). All of the trucks and their respective seats were considered to be in good working order during the study. Measurement periods for the truck groups had similarities, but varied from 2 to 58 minutes. Sampling times for the older trucks included a mean of 19.5 minutes and a standard deviation (STD) of 6.5 minutes compared to a mean of 40.8 minutes and a STD of 12.1 minutes for the newer trucks. Data collection coincided with the approximate delivery and first operation of the new trucks, and occurred approximately 12 months apart under similar weather and road conditions, and with the same drivers except an additional driver was included with the older trucks. Truck routes were somewhat different in that quarry production had changed location in the time between data collection activities. Overall, the results suggest that the newer trucks may provide better overall isolation to drivers/operators from WBV exposure compared to the older trucks operating at the quarry; although, this will need to be confirmed with additional measurements. Considering the higher variability and shorter sampling times for the older trucks, the results should be viewed with caution. For two of seven trials, the older trucks showed that seats amplified vibration, i.e., a transmissibility (T) >1.0. Seat T for the older trucks ranged from 0.31 to 1.17 with a mean of 0.77 and STD of 0.32. This contrasted with the newer haulage trucks where seats amplified vibration in 3 of 8 trials. In this case, T did not vary greatly and ranged from 0.87 to 1.05 with a mean of 0.97 and STD of 0.07. Regarding older trucks, in five of seven trials, the seat (output) data of weighted root-mean square (RMS) acceleration (wRMSZ) for the dominant z-axis exceeded the action level of 0.5 m/s2 action level recommended by the European Union Good Practice Guide for WBV (EUGPG) and levels exceeded the recommended exposure limit of 1.15 m/s2 in two of the seven trials. The wRMSZ values for the older trucks varied from 0.41 to 1.83 m/s2 with a mean of 0.99 and STD of 0.57. Similarly, newer trucks indicated a narrower range of wRMSZ from 0.38 to 0.95 m/s2. The mean wRMSZ was lower for the newer trucks at 0.58 m/s2 with a STD of 0.23 m/s2. Similarly, newer trucks indicated wRMSZ reached or exceeded the action level in four of eight trials. None of the trials with the new trucks showed wRMSZ levels that reached or exceeded the recommended 1.15 m/s2 exposure limit. As an indicator of driver/operator discomfort, overall weighted total RMS acceleration (vector sum) values seem to show a ?rougher? ride for the older trucks. The vector sum values for these trucks ranged widely from 0.70 to 2.59 m/s2 and, in four of seven trials, showed levels greater than 1.40 m/s2. The mean vector sum was 1.44 m/s2 with a STD of 0.75 m/s2. Comparatively, the newer trucks exhibited less variation with a range from 0.69 to 1.59 m/s2. The mean vector sum was 1.02 m/s2 with a STD of 0.35 m/s2. Vibration dose values for the dominant z-axis (VDVZ), gave a sense of vehicle jarring/jolting conditions. All trials with the older trucks were within the recommended EUGPG action level of 9.1 m/s1.75. On the other hand, in three of eight trials, both newer trucks exceeded this action level with values of 9.18, 12.58, and 13.21 m/s1.75. Neither truck group showed VDVZ that exceeded the exposure limit of 21 m/s1.75. A statistical analysis was not conducted, since the differences reported between truck groups may not be statistically significant owing to the relatively small sample size. Road conditions, changes in the truck routes, and driver/operator differences (e.g., stopping and turning) are possible factors in the higher VDV for the newer trucks.

Physiol. 11): L365–L374, 1994.—Although integrity of the alveolar basement membrane may influence progression of lung injury induced by inhaled particulates, little is known about direct effects of coal dusts on the alveolar epithelium or its extracellu¬lar matrix (ECM). Effects of dust on synthesis of cell and ECM proteins by type II cells (T2P) was thus investigated. Three coal dusts (anthracite no. 867; bituminous no. 1451 and no. 1361) and mine dust MIT-3, of respirable size, were studied as a function of dose and time over 3 days of primary T2P culture. On day 1, 750 µg/ml of 867 were required to increase relative synthesis of ECM proteins (ECM/cell). MIT-3, 1451, and 1361 were without effect. By day 3, 867 or MIT-3 increased ECM/ cell 60-100% at 300 µg/ml; 1451 produced modest, dose-dependent stimulation, whereas 1361 remained without effect. None of the dusts caused significant cytotoxicity. The results show dose- and time-dependent effects of well-characterized coal and mine dusts to modify partitioning of newly synthesized proteins into the ECM and suggest that coal dust exposure may modulate structure or function of the subepithelial basement membrane.basement membrane; cell culture; inorganic dust; lung injury; extracellular matrix deposition; pulmonary epitheliumCHRONIC EXPOSURE to coal dust may be associated with development of pulmonary fibrosis and consequent loss of normal lung function (see Ref. 26 for review). Al¬though the pathways of inorganic dust-induced lung injury are recognized to involve an inflammatory re¬sponse, the underlying pathogenic mechanisms are not understood in detail. Acute and chronic responses to dust exposure involve a variety of interacting cells of the immune system, including macrophages, T cells, and B cells, along with pulmonary fibroblasts. These responses reflect direct effects of the dust particles, as well as indirect effects mediated by soluble factors, including cytokines and eicosanoids (see Ref. 41 for review).Studies of pulmonary macrophages and fibroblasts following silica exposure suggest that dust-induced damage at the airway surface may involve cellular injury through oxidant-mediated pathways. Coal dusts and related inorganic particulates can activate the alveolar macrophage (42) to produce fibronectin (15) and cytokines, such as interleukin-1 or tumor necrosis factor (4) and/or other soluble products which act locally (29) to cause acute cellular injury and to initiate the progression of fibrotic lung damage (16). Type I epithelial cells are typically more susceptible than type II cells to oxidant injury. Type I cell damage and subsequent death leads to denudation of the subepithelial basement membrane (2, 21) and can be associated with leakage of plasma constituents from the pulmonary vasculature into the alveolar compartment (13)."

High-pressure shrouded sprays were first tested on a bench to determine air-moving capability and dust collection efficiency. The bench-scale tests indicated that at 500 psi and above, the dust collection efficiency, when sampled downstream of a water eliminator, was about 99 pet for two types of nozzles. However, for the same water flow, the induced airflow was about three times greater in one type than in another. Since the only difference in the nozzles was the spray angle, this factor was important in air movement through a shrouded spray. Full-scale model mine tests were then conducted to determine the reduction in dust concentration behind the line curtain and at the operator position produced by high-pressure sprays in a shroud. The shroud was mounted on top of the boom of a model continuous miner, and high-pressure (greater than 500 psi) sprays were installed inside the shroud. The inlet of dusty air to the shroud was open toward the face and the outlet toward the exhaust line curtain. The high-pressure sprays were compared with conventional sprays mounted at the head of the continuous miner and directing water toward the face. The difference at the operator position (opposite side of brattice) was significant in that, under certain conditions, with conventional sprays an increase in respirable dust concentration of 471 pet was noticed compared with dry operation due to rollback from face. High-pressure shrouded sprays, on the other hand, decreased the dust concentrations at the operator position (opposite side of brattice) by 28.2 pct.

In safety work a study of past accidents is of value in determining means of reducing future accidents of the same character. This is particularly true with reference to coal-mine explosions. Data from publications of the United States Bureau of Mines and from published reports of the State inspector of coal mines of Colorado are presented in this circular in the belief that they will be of value to operators by indicating methods of preventing future coal-mine explosions. The tables included are complete only with regard to major disasters. The Bureau of Mines considers as a major disaster one in which five &apos;or more men are killed. However, this circular also includes a considerable number of minor disasters, Various explosions in the coal mines of Colorado from 1883 to 1932, inclusive, are listed with information available as to their probable cause, number of men killed and measures which, if they had been in use at that time, might have prevented them. John McNeil, first Colorado State coal-mine inspector, on page 43 of his first report for the period ended July 31, 1884, made the following recommendations for the prevention of explosions in the coal industry: True causes could be traced either to insufficient ventilation, carelessness and lack of rigid discipline in carrying on energetic ventilation and other precautions needed to be enforced in a mine giving off explosive gases. *** When any quantity of fine coal dust is observed in an entry, air course, or room it should be made damp and be removed from the mine, and thus beep every place as free from dust as possible. *** A system of bratticing close up to the face of each and every place in the mine from crosscut last made (whether gas should just then show its presence or not) which should not be more than 50 or 75 feet according to the nature of the mine.

A study of the high magnification of some inorganic fibers which are found in nature or produced in the laboratory, has been conducted with the electron microscope. This series of electron microqraphs shows the important morphological differences exhibited by potassium-lead silicate, lead-aluminum silicate, chrysotile, wollastonite, silicon nitride, potassium titanate, cobalt chrysotile, magnesio-fluor-richterite, fluor-richterite, refractory oxide fiber, silicon carbide, massive serpentine, and palygorskite. A ribbon or a thin lath 250-2500 A. wide is the structural form of potassium-lead silicate and lead-aluminum silicate. Chrysotile, cobalt chrysotile, and massive serpentine all have a tubular structure and a fairly uniform (150-300 A.) diameter. Wollastonite, potassium titanate, fluor-richterite, and magnesio-richterite crystals have a lath shape, and potassium titanate laths are laminated ribbons. Single fibers of silicon nitride and palygorskite are ribbons, and the silicon nitride sample contains many twins (branching fibers). Palyqorskite fibers are highly flexible and have cross-sectional measurements of 150-300 A. Silicon carbide fibers differ from other fibers because their structure consists of several different forms. The sample used in this study had, predominately, a margarite-type structure. The refractory oxide fiber lost, all its apparent fibrosity after being prepared for electron microscopic examination, and only irregular fragments are visible in the electron micrographs.

Multi Mineral Corporation conducted a mining research program at the United StAtes Department of the Interior&apos;s Bureau vi Mines Oil Shale Mining Environmental Research Facility in the Central Piceance Creek Basin, Rio Blanco County, Colorado. The program consisted of mining, environmental and geotechnical research data and. the results are described in this report.

As recently as 1915, helium was available only in very small quantities at a cost equivalent to $2,500 per cubic foot. It was a laboratory curiocity, and its properties were of interest to a limited number of scientists (11).3/ During the past 30 years, however, helium has grown from a plaything of scientists to a factor of importance in the prosecution of the recent war. Navy blimps filled with nonflammable helium proved to be more than a match for German submarines operating in the Atlantic. American bombers were able to fly unheard-of distances to drop bombs on Tokyo, Nagoya, Yokahama, and other Japanese cities because they had been made lighter with magnesium, arc-welded in a helium atmosphere. The Army utilized helium-filled radio-sonde balloons to obtain vital information concerning weather conditions affecting D-day invasions and other military maneuvers.

Mining haul trucks comprise the majority of the equipment used in underground limestone mining operations and are known to emit high levels of noise. A previous study conducted by the National Institute for Occupational Safety and Health (NIOSH) indicates that 70-90 percent of all miners have a noise-induced hearing loss (NIHL) great enough to be classified as a hearing disability by retirement age. These results demonstrate the public health need to protect the hearing of workers in the mining industry, including haul truck drivers. Cab enclosures present an opportunity to isolate the haul truck operator from both truck and other noise in the mining environment. A total of twenty-five haul truck cabs were studied which were divided into three style (treatment) categories determined by soundproofing features and technology for noise reduction: old-, new-, and retrofitted-style. This study examines the contribution of cab acoustics, operator performance, and maintenance to noise reduction for each cab style. Dosimeters were used to measure 8-hr time weighted average sound pressure levels (TWA8 SPLs) inside and outside the cabs to determine if different acoustical treatments affect cab attenuation. Adherence to the Mine Safety and Health Administration (MSHA) Permissible Exposure Limit (PEL) of 90 dB TWA8 (with a 90 dB threshold) was used as the main indicator of overall noise reduction achieved. Dosimetery results indicate only 2% of the samples exceeded the PEL, but samples could still be reduced much further. Descriptive and comparative statistics indicate that noise levels inside the new-style cabs are significantly lower than the other two cab styles. Also, data suggest that there is no difference in noise exposures when comparing the old-style to retrofitted cab styles. Operator influence (opening doors and windows) was a significant factor for increasing noise exposure. This paper demonstrates that properly designed cabs can achieve major noise reductions, but noise levels could still be reduced much further below the MSHA PEL. New-style cabs, equipped with modern noise-reduction treatments, exhibit much lower noise exposures than the other two cabs styles, and the effectiveness of the current noise-reduction treatments for retrofitted cabs is questionable. Haul truck driver observations indicate that improved noise exposure reduction training is needed. Finally, specific targets for future noise reduction research are suggested that will further contribute to the prevention of hearing loss for haul truck operators.

Distillable oils derived from coal were desulfurized and hydrogenated at 2,500 psig in a two-step vapor-phase catalytic operation to give a product rich in saturated cyclic hydrocarbons. The first step was mainly a desulfurization at 400 C, the second a saturation at 300 C. One of the oils used as feed stock was from low-temperature carbonization of a bituminous coal, the other was from the liquid-phase hydrogenation of coal in the Bureau&apos;s pilot plant. The product from the saturation step was distilled to remove the light ends and high-boiling residue and to recover a fraction having acceptable properties with respect to heating value, density, freezing point, viscosity, and boiling range according to specifications suggested by the Air Force for ultra-high-speed aircraft fuel. Quantities of the fuel fraction available were too small for the thermal stability test approved by the Air Force. The experiments, performed on laboratory scale, comprised mostly a study of the effect of varying the oil feed rate (at constant oil-to-hydrogen ratio) on the yield of the desired fuel fraction. The yield of the fuel fraction made from low-temperature carbonization oil was a little more than 1 percent of the weight of coal carbonized; that from coal-hydrogenation oil was about 12 percent of the weight of coal hydrogenated. A few attempts to do both desulfurization and saturation in one operation with sulfur resistant catalysts, developed for vapor-phase hydrogenation or hydroforming, were unsuccessful. By leaving the low-boiling ends in both feed stocks to desulfurization, another product was obtained: a low-sulfur aromatic cut boiling below 200 C. This material would be suitable as a solvent or a chemical raw material.

"INTRODUCTION In the manufacture, storage, transportation, and use of combustible gases and vapors, explosion hazards, which at times may be present when air becomes mixed with the gases and vapors, become of primary importance. Such conditions are especially dangerous where large volumes of gaseous atmosphere are involved, because an explosion under such circumstances is capable of doing tremendous damage to both property and personnel.Because of its interest in safety in the mining and allied industries, the Federal Bureau of Mines has investigated, and published results dealing with, the explosibility of various types of combutible atmospheres. The explosion hazards of natural gases have received attention, and results giving the limits of inflammability and ignition temperatures in air have been determined and published. 4/ Other investigations have shown how explosions of natural gas-air mixtures may be prevented by the addition of nitrogen and carbon dioxide. 5/Because of the increased demand for crude oil for war purposes, the application of secondary-recovery methods of stimulating oil production have received considerable attention during the last few years. The process consists in the injection of natural gas into partly depleted oil wells to cause the oil in the sands to flow into producing wells and so increase the amount of recoverable oil. 6/"

The direct method test was developed for general mine safety applications by the U.S. Bureau of Mines in the early 1970&apos;s to measure the gas content of coal samples obtained from the relatively shallow depths of eastern U.S. coal mines. Over time, applications for gas content data evolved which required increasingly accurate desorption values. It became evident that some of the inherent weaknesses in the original direct method test could best be addressed by abandoning the water displacement based desorbed gas volume measurements. The ideal gas law based modified direct method (MDM) and associated desorbed gas volume measurement apparatus was developed to provide the most accurate desorption values possible for rock samples obtained from any gas-bearing strata. The MDM has a number of significant advantages, including: (1) increased accuracy, especially for low gas content samples, (2) a test apparatus with an integral sampling port to facilitate the collection of uncontaminated gas samples, (3) an inherent head space effect correction in the periodic MDM desorbed gas volume calculations, (4) measurement of positive and negative desorbed gas volumes, (5) elimination of differential gas solubility effects as a source of error inherent in the water displacement methods, and (6) a compact and rugged test apparatus that along with the elimination of the ?glassware? required for the water displacement methods, facilitates its use in the field.

The National Institute for Occupational Safety and Health (NIOSH) and the Mine Safety and Health Administration (MSHA) conducted a joint survey to determine the range of coal particle sizes found in dust samples collected from intake airways of US coal mines. The last comprehensive survey of this type was performed in the 1920s. The size of the coal dust is relevant to the amount of rock dust required to inert the coal dust, with more rock dust needed to inert finer sizes of coal dust. Dust samples were collected by MSHA inspectors from several mines in each of MSHA&apos;s 10 bituminous Coal Mine Safety and Health Districts. Samples were normally collected in several intakes at each mine. The laboratory analysis procedures included acid leaching of the sample to remove the limestone rock dust, sonic sieving to determine the dust size, and low-temperature ashing of the sieved fractions to correct for any remaining incombustible matter. The results indicate that particle sizes of mine coal dust in intake airways are finer than those measured in the 1920s. This finer size coal dust in intake airways would require more incombustible matter to be effectively inerted than the 65% incombustible specified in current regulations.

The investigation of the nickel - gadolinium phase diagram was undertaken to determine the alloying behavior of these two metals . The data from meltingpoint determinations , thermal analyses , metallographic examinations , and X- ray diffraction analyses revealed a phase diagram with three eutectic points and seven intermetallic compounds . Very limited solubility of the phases in each other was observed . The five phases , Ni Gd , Ni , Gd , Ni Gd , Ni, Gd , and NiGd¸ , melted incongruently at about 1,300 ° , 1,270 ° , 1,180 ° , 1,070 ° , and 775 ° C , respectively . Ni Gd and NiGd melted congruently at approximately 1,550 ° and 980 ° C , respectively . Four of the compounds were determined to crystallize in the hexagonal system : Ni Gd , Ni Gd , Ni , Gd , and Ni Gd . Ni Gd was determined to be cubic and NiGd , orthorhombic . The crystal system of NiGd was not determined . The eutectic points occurred at 13 , 67 , and 85 wt pct gadolinium , and the isotherms at 1,290 ° , 890 ° , and 700 ° C , respectively .

The fibrogenic effects of silica in experimental animals and humans has been extensively studied (I 7, 20). However, the pathogenesis and molecular mechanisms underlying lung injury and fibrosis resulting from silica exposure remain to be elucidated. It is well-documented that silica can stimulate monocyte-macrophage lineage cells to produce cytokines and growth factors, including interleukin-I (IL- I) (20), tumor necrosis factor-ex (TNF-cx) (I 7), insulin-like growth factor-I (IGF-1) (4), and transforming growth factor-P (TGF-P) (24). Consequently, it has been suggested that the fibrogenic effect of silica may be mediated by cytokines in the fater stages of silicosis. During the early phase of interaction between silica and the pulmonary alveolar macrophage, several direct and indirect lines of evidence suggest that reactive oxygen intermediates (ROI) may play a pivotal role in silica- induced tissue damage (2I, 22). One of the most important components of and contributors to ROI is nitric oxide (NO) produced from L-arginine and molecular oxygen under the catalytic influence of NO synthase (NOS) (I 9). At present, at least three NOS genes have been cloned: a constitutive neuron NOS, a constitutive endothelial cell NOS and an inducible NOS (iNOS) (16). A number of factors have been shown to differentially induce iNOS gene expression in different cell lines including bacterial cell products and foreign antigens. During inflammation, NO is a double-edged sword in terms of the host immune defense mechanisms and the physiologic response to an inflammatory insult. The macrophage is one of the major cellular sources of NO. During the early phase of acute inflammation, NO produced from the macrophage appears to facilitate the clearance of pathogens and promotes a local relaxation effect on the microcirculation of vascular smooth muscle. When the production of NO is left unattenuated, especially under oxidative stress, direct cytotoxic effects of NO can emerge through the formation of peroxynitrite as a result of a reaction between NO and superoxide (2).
