The mucus secreted in the respiratory tract provides the first barrier against inhaled particulate and gaseous toxicants. The major component of mucus which is responsible for the physical properties and protective functions of the respiratory mucus is mucin. We have initiated investigations to determine the effects of coal mine dust on the synthesis of respiratory mucin by rat trachea under in vitro and in vivo conditions. In this study we have established optimum conditions-:ro-r~ the asceptic growth and mucin secretion of rat tracheal explants in culture. The best outgrowth of ciliated epithelial cells and maximum mucin production was obtained in CMRL 1066 medium containing 10% fetal calf serum. The explants continued to secrete mucin even after three weeks in culture. For the estimation and characterization of mucins the tracheae were cultured in the presence of [3H] glucosamine and [35S] sulfate or [14C]leucine. The mucins were isolated from the medium by gel filtration, treatment with hyaluronidase, ion exchange chromatography and CsBr density gradient centrifugation. Precipitation of polyanionic material in the culture media with cetyl pyridinium chloride and fractional extraction with NaCl also yielded mucin free of glycosaminoglycans, but in low yields. The tritium label in the purified mucin was distributed in sialic acid (50%) galactosamine (37%) and glucosamaine (13%). The density of purified mucin as determined by CsBr gradient centrifugation was about l.43g per ml in comparison to l.46g per ml for human respiratory mucin with a carbohydrate content of about 80% by weight. Alkaline borohydride treatment released saccharides which contained about 80% of the radioactivity. The effect of adding varying amounts of coal dust to the culture medium is under investigation. Coal is a combustible, carbonaceous, sedimentary rock that is formed by the accumulation, compaction, and physical and chemical alteration of vegetation [Simon and Hopkins 1981; Bates and Jackson 1987]. Coal is classified according to its type, grade, and rank. The type of coal relates to the plant materials from which the coal originated. The grade of coal refers to the purity of the coal-or the amount of inorganic material (including ash and sulfur) left after the coal is burned [Bates and Jackson 1987; Stefanko 1983]. The rank of coal indicates its degree of metamorphosis and roughly correlates to the geological age of the coal or the geological environment from which it has been mined [Bates and Jackson 1987]. Rank also indicates the percentage of carbon in dry, mineral-free coal [Whitten and Brooks 1973] and the degree to which the coalification process has progressed [Larsen 1981]. The geological process of coalification begins with organic materials (e.g., celluloses, lignins, and other plant compounds that are deoxygenated and then dehydrogenated) and ends with coal of various geological ages, from lignite to anthracite [Larsen 1981]. Table 3- 1 presents the American Society for Testing and Materials (ASTM) classification of coals by rank. According to Parkes [1982], high-rank coal includes anthracite and semianthracite coal ("hard coal," with 91% to 95% carbon); intermediate-rank coal includes low-, medium-, and high-volatile bituminous and sub-bituminous coal ("soft coal," with 76% to 90% carbon); and low-rank coal includes lignite (with 65% to 75% carbon or less). The rank of coal tends to increase from the western to the eastern United States, with anthracite occurring primarily in eastern Pennsylvania [Schlick and Fannick 1971]. Most of the coal currently mined in the United States is bituminous [Given 1984]. A classification of the coal in the United States is provided in Table 3-2.

The temperature entropy (T, S) chart? of thermodynamic properties of nitrogen herewith presented is a complete (1949) revision and a considerable extension of similar charts prepared by the author for use of the helium section of the Bureau of Mines about 25 years previously on the basis of inadequate data then obtaining. During the interval since that early effort, much experimentally determined information on nitrogen properties and behavior has been published, which now permits a more nearly accurate layout and more extensive charting of these relations.

As part of the U.S. Bureau of Mines (USBM) program to characterize subsidence this combination of software and hardware has been used to remotely monitor overburden response to longwall mining. Background The principle of time domain reflectometry (TDR) is being used by the USBM to monitor strata displacements induced by mining. This capability has been enhanced by the availability of cable testers with serial communication modules that allow users to get TDR records in digital form. Actual field installations involve coaxial cables that arc hundreds of meters long, and the many records obtained when monitoring such long cables on a daily basis motivated development of software to view interactively and compare waveforms and to analyze signature changes. Time domain reflectometry (TDR) was developed in the 1960's as a method to locate discontinuities in coaxial transmission cables. The concept has been extended to measurement of properties of materials into which probes are embedded, such as water content of soil, and evaluation of coaxial cable dielectric behavior. This technique was adapted by the USBM in 1975 for use in checking the movement of rock during underground mining. Coaxial cables are grouted into a rock mass that is expected to cave, and progressive rock movement causes the cable to deform. Consequently, there is a progressive change in cable electrical properties that produces changes in TDR pulse reflection signatures. TDR technology has been advanced by Northwestern University to the point where it is now possible to distinguish shear deformation from tensile deformation, and to quantify rock mass displacements, by analyzing these changes in TDR signatures.

The Location Group conversed in the Petroleum Room of the Green Center with the purpose of addressing the following list of topics in light of the Bureau of Mines' requirements for detecting and locating trapped miners by electromagnetic methods. These topics include: Noise, Sources, Antennas (transmit and receive), the Propagation Medium, Signal Processing, Optimum Frequency, Receivers, Pulse vs. CW Techniques, Passive Location Techniques, and Obstacles. Practical constraints such as equipment size, weight and power requirements, cost and intrinsic safety were kept in mind during these discussions. It was emphasized that in-mine equipment must be inexpensive, rugged, simple and lightweight. Otherwise, the mine operators probably would not buy it nor would the miners be able to effectively use it. Present techniques for experimentally detecting and locating trapped miners were discussed, along with techniques to improve their location accuracy. Analytical, numerical, and scale modelling methods were considered as a means of giving further insight into measurement, anomalies caused by terrain variations, conductivity contrasts, and obstacles. After discussion of these topics, conclusions were drawn and existing problem areas were identified. These problem areas were addressed in developing guidelines for future efforts. Following the discussion period, the Chairman gave a summary of the Location Group report and comments were solicited from the entire group. 2.0 GROUP DISCUSSION HIGHLIGHTS 2.1 Noise W. Bensema asked the group whether the noise data provided by NBS was adequate. It was determined that for location systems analysis, the most important form of noise data was the frequency spectra of vertical and horizontal magnetic fields on the surface, and the answer to his question was affirmative. Drs. Wait and Frischknecht pointed out the difficulty in accurately measuring the true vertical magnetic field since these measurements are heavily influenced by the loop setting and the local geology. The Bureau of Mines in its investigations of respiratory apparatus for use in mines and allied industries, tests equipment that is voluntarily submitted by manufacturers in accordance with published schedules of tests. These tests are designed to insure that the equipment has the minimum require- ments for safety in use. Approval certificates are issued for apparatus that meets the bureau's specifications, and the memfacturer is permitted to mark his equipment with a seal showing that it has been approved as "Permissible." A complete list of permissible self-contained oxygen breathing apparatus, gas masks, and hose masks tested before July 1, 1930, was published in Bureau of Mines Information Circular 6362.1 The present list includes all equipment tested and approved up to and including April 1, 1931.

Mountain bumps have been a problem in the coal-mining industry since the 1920's. The problem became severe in the late 40's and early 50's with the introduction of mechanized mining. Fatalities and serious injuries resulted from mountain bumps during this period. Mountain bumps again caused fatalities in the 80 's. Interviews were conducted mostly with retired coal miners having experience in bump conditions with five different coal companies. These men held positions as president, general manager, roof-control specialist, mining engineer, inspector, superintendent, mine foreman, shift foreman, section foreman, miner operator, electrician, and general laborer. Together, their comments give a good idea of the things tried to mitigate mountain bumps in the Appalachian Coal Fields.

The Bureau of Mines investigated an electrolytic technique for oxidizing ore slurried with brine to determine its effectiveness in extracting mercury from ores. Optimization experiments conducted on a 2.5-pound-mercury-per-ton ore resulted in 95 percent mercury extraction with operating conditions of 30° C, a current density of 0.5 ampere per square inch, and a treatment time of 3.5 hours. Corresponding power consumption amounted to 38 kilowatt-hours per ton of ore treated. Amenability experiments conducted on 62 different mercury ores resulted in 90 to 99 percent mercury dissolution from ores containing 0.6 to 20 pounds of mercury per ton of ore, Treatment time ranged from 1 to 7 hours in 4-to 20-percent sodium chloride solutions, and power consumption was in the range of 10 to 50 kilowatt-hours per ton of ore. Pilot mill experiments in a 100-to 200-pound-per-hour extraction plant have been in progress to quantify power and reagent requirements. These data will be reported in a subsequent publication.

Supernatant media of dust-exposed pulmonary alveolar macrophages (PAMs) were inactive in assays for both Interleukin-1 and fibroblast growth factors (FGF). We have begun to evaluate several factors that may interfere with dust-PAM interactions. To determine the effect of sterilization on the activity of dusts, PAMs were exposed to autoclaved dust, heat-sterilized dust or to dust that had not been heated. Supernatants from the first two groups were inactive in the FGF assay, but supernatant from PAMs exposed to unheated dusts stimulated growth of lung fibroblasts. Recent data have revealed that freshly crushed mineral dusts possess labile free radicals that are absent in dust that has been stored for more than a few days. Suspensions .were prepared of a "stale" sample of anthracite dust 867 and of a freshly ground sample of the same dust. These suspensions were instilled intratracheally into guinea pigs under general anesthesia. Two, five or eight days later, PAMs were collected from the lungs by bronchoalveolar lavage, and the cells were counted. At two and five days after instillation, all lavage suspensions contained 70 to 85% PAMs, of which 5 to 18% contained phagocytized dust particles. On day eight there were again 80-84% PAMs in all suspensions, but in the presence of "fresh'" dust, 48% of PAMs. had phagocytized particles in comparison to 16% in the presence of '"stale'" dust. A similar experiment was performed in short-term cell culture. During 24 hours, >95%. of PAMs phagocytized dust particles, whether or not the dust was "stale" or '"fresh". Our studies are being extended to determine the effect of (1) removing surface oil contaminants by organic extraction and (2) suspending dusts without surfactant.

Knowledge of the water content of oil-bearing formations is important in petroleum-reservoir evaluation work. Electric logging provides a means for making reliable estimates of water content if resistivity-saturation relationships representative of the formations are available. Relationships can be determined in the laboratory by a number of methods. This report describes and presents results of a laboratory investigation conducted at the Bureau of Mines Appalachian Experiment Station, Morgantown, W. Va. (1) to evaluate an evaporation method of attaining water saturations in sandstone samples when determining resistivity-saturation relationships and (2) to compare saturation exponents derived from data obtained, using different methods of achieving water saturations. A series of tests was conducted to determine the effect of evaporation on salinity distribution within the core specimen. Results of these tests show that a zone of high salinity was set up at the surface of a sample during evaporation and that long periods were required for the sample to reach resistance equilibrium after evaporation had ceased. Equilibrium time ranged from several minutes to several days, depending on the sample size and the technique used. The high-salinity zone was removed from the sample by using auxiliary porous material in capillary contact with the sample surface during evaporation. Salinity determinations and resistance measurements indicate that, at saturations greater than the minimum interstitial values, the mechanism of flow during evaporation was one of moisture migration mainly as a liquid from the interior of the test specimens to their exterior surfaces. The concentration of the contained brine remained essentially unchanged, except for a narrow zone near the evaporating surfaces.

The U.S. Bureau of Mines has developed an acoustic test for determining if a rock salt sample is from a "normal" nongassy salt face or from a gassy face where an outburst has the potential to occur. The test is based on the observation that a salt sample taken near an outburst-prone zone produces an audible popping sound when it is dissolved in water. The sound is produced when pressurized gas that is trapped in salt samples is released. In the acoustic test, the sound is electronically measured, digitized, and transmitted to a microcomputer; the output is an average decibel value that classifies the sample as "normal" or "outburst." The acoustic test procedure and equipment are described, and raw test results and data analysis are shown. Results are also shown for several salt samples that were analyzed using a laser Raman microprobe.

During the 1970?s, the Bureau of Mines investigated a particle agglomeration technique for improving the flow of leaching solution through heaps of clayey or crushed, low-grade gold-silver ores, wastes, and tailings. This technology has been adopted on a broad scale by the precious-metals-processing industry. This report presents information on five commercial operations that have benefited from agglomeration technology and that represent a cross section of current heap leaching practice. The technology is cost effective because of decreased leach times and improved precious metal recoveries.

The U.S. Bureau of Mines is currently investigating the use of genetic algorithms (GA's) for solving optimization problems. This computer search technique, based on the mechanics of natural genetics, was used to perform simple least squares curve fitting. Three examples are presented in which a GA manipulates binary coded strings to produce near-optimal solutions to least squares curve-fitting problems after having viewed only a small portion of the search space. The examples include fitting data to the equation of a line, the equation of a parabola, and the Ree-Eyring equation.

It is of interest to know how to obtain as high an absorption efficiency as possible from a CO2-absorption canister used in closed-circuit breathing apparatus. In this study the Bureau of Mines found that varying the bed depth and the cross-sectional area of a canister changes its absorption characteristics. In tests using four ,1 rectangular, variable-depth canisters with different cross-sectional areas, the Bureau found that, given a constant weight of the chemical absorbent, LiOH, and a constant COa input, the canisters with a greater cross-sectional area reached C02 levels of 0.5 volume-percent in the effluent gas sooner than did the canisters with a greater bed depth. However, resistance to flow through the canister increases with bed depth, and a trade-off is inevitable. It was anticipated that the efficiency of a canister would increase with increasing intergranular air space approaching the tidal volume of the breathing waveform and then level off at some point under 100 percent. This postulation was partially substantiated. Cylindrical and radial canisters were also tested and compared. It appears that shape is not as important a factor in absorption efficiency as bed depth, since neither canister differed significantly from the rectangular canisters with regard to absorption efficiency values.

The approval of the U. S. Bureau of Mines when granted to a manufacturer for a gas mask is a certification that the particular device has been submitted to the Bureau of Mines for inspection and test and has been found to meet that bureau's published minimum requirements for safety , durability , and satisfactory performance . Any de- Gas masks are approved by the Bureau of Mines only as a complete unit or assembly of parts , as canister , facepiece and harness . This is in order that responsibility for rigid adherence to the bureau's requirements may be definitely located and not divided . vice marketed under this approval must be a unit composed of parts identical in detail of design , assembly , workmanship , and quality , and produced under the same control and inspection as the original device to which approval has been granted . The particular parts which comprise the approved assembly are designated in a letter to the manufacturer at the time the approval is granted . In recent years they are also specified in the approval plate authorized for use in connection with the sale of the device . Each part permitted in the assembly is legibly marked by name or number for the guidance of users of masks in ordering and identifying approved parts when making repairs , and thereby maintaining their equipment as an approved device . In view of the foregoing , any change in the design of parts or deviation from the permitted assembly of parts would violate the conditions under which approval was granted and the device would not be considered as meeting the approval requirements of the Bureau of Mines . The U. S. Bureau of Mines has been frequently requested to interpret the approval status of a gas mask assembled from parts which have separately met its requirements as components of two or more approved devices but which in the form of a complete unit has not been submitted for tests or been granted approval . Responsibility for the elements and for their proper assembly is in such case divided . Such assembled units have no status as permissible equipment under the Bureau of Mines system of schedules . The reasons underlying this policy are that the approval is granted only to some one who assumes responsibility for the whole and that the details of assembly and coordination of parts are considered to be important to the satisfactory performance and safety of the device .

NIOSH has developed an automated PC-based seismic event (induced earthquake) monitoring system for use in mine ground control studies. The distribution and coordination of data acquisition and processing functions across a local area network is a central feature of the system. Mine-wide installations utilize multiple, autonomous seismic networks located both underground and on the surface. Seismic event locations, magnitude estimates, and other processed data are distributed for display and analysis on network nodes both underground and in the offices of mine engineers and managers. This paper describes the system’s design philosophy, hardware and software components, examples of installations, and a few observational results.

Four catalytic converters for diesel exhaust systems were tested to determine their effectiveness in reducing the amount of harmful substances discharged into the air. Tests were conducted with four different catalytic converter designs, using two diesel engines as exhaust gas generators. (Of the four noble metal catalytic converters, three are commercially available.) Testing showed that the catalysts varied widely in oxidation efficiency at light to intermediate engine loads, but that all were very effective in reducing emissions of CO, HC, aldehydes, and odor intensity at heavy loads (engine operating modes resulting in high exhaust temperatures). Particulate loading, particulate size, and NOx emissions were generally unaffected by any of the catalysts. The catalysts had one ad-verse effect on exhaust quality--increased sulfate emissions. Because of the potential problem of sulfate emissions, the exclusive use of catalytic reactors for emissions reduction does not appear to be satisfactory. However, when used with a means for subsequent removal of sulfates, a catalyst can function as a key component in an emissions control system for diesel engines operated in underground mines.
