"A sensitive colorimetric assay for detecting mucins and glycoproteins rich in 0-glycosidically linked saccharides is reported. The method combines the susceptibility of N-acetylgalactosamine terminating 0-glyco idically linked saccharides to P-elimination with the Morgan-Elson reaction for N-acetylhexosamines with free reducing ends. All mucin and mucin-type glycoproteins but none of the serum-type glycoproteins tested resulted in characteristic color production. All mucins tested gave linear responses in the range 5 to 200 ""g and the assay was also adapted to the microscale involving the use of 96-well microtiter plates. The microassay in which the volumes of samples and reagents are scaled down 2.5-fold was particularly useful in monitoring of mucins, in the presence of other glycoconjugates, in large numbers of samples obtained during fractionation procedures. Cesium chloride, cesium bromide, potassium thiocyanate, and various detergents do not interfere with the colorimetric determination. Guanidine hydrochloride, cesium triftuoroacetate, and B-mercap- . toethanol decreased color by 30 to 45%; however, the interference was not serious to prevent the use of the method for detection of mucins in their presence. The use of the method for the specific detection of mucin during fractionation by gel filtration and density gradient centrifugation of cystic fibrosis sputum samples is demonstrated. Mucins are an important class of biomacromolecules whose structure-function relationship is poorly understood (1,2). The investigations of mucins are hampered by difficulties in purifying these very large size molecules which appear to avidly bind other components of the mucus (3,4). One of the problems during purification is the specific detection of mucins. For example, during the various purification steps, the fractions containing mucins are typically detected by assaying for neutral sugars (hexoses) (5,6), for sialic acids (7), or for periodate oxidizable saccharide units (8). However, these methods are not entirely satisfactory because neutral sugars, sialic acids, and periodate-oxidizable saccharides are also components of other glycoconjugates. In addition, the neutral sugar and sialic acid measurements are also subject to interference from high levels of proteins and nucleic acids."

"INTRODUCTION Experience in drilling oil and gas wells has shown that the voidance of blow-outs, twist-offs, fishing jobs, loss of circulation, and heaving and caving of shale formations into the drill hole depends largely on how well the drilling-mud fluid serves its purpose. A possible method for the control of the drilling fluids may be seen from a study of the relation between the particle-size distribution and some of the important drilling-fluid properties, such as the ability to build quickly a relatively impervious mud sheath on the wall of the hole in the porous formations penetrated by the bit during drilling operations. In a wartime economy, which urgently requires conservation of steel, proper functioning of the drilling fluid is particularly important because, as Parks 4/ has stated, intelligent use of good drilling fluids makes possible drilling of wells for thousands of feet in open holes without the necessity of setting protective strings of casing to control the movement of salt water, gas, and troublesome formations into the borehole; consequently, especially in deep wells, of proper drilling fluids materially reduces the need for casing and lowers the cost of drilling and completing the wells."

The Bureau of Mines developed a flotation method for concentrating spiral tailing produced from pilot plant tests on the Champion mine ore fines . The feed material contained about 49 percent Fe and 15 percent SiO2 and was posed mostly of specular hematite , muscovite , and chlorite . Bench- scale , batch flotation research developed a reagent suite composed of tall oil - fatty acid , fuel oil , sulfuric acid , sodium fluoride , and sodium silicate for recovery of iron minerals in the froth product . Concentrate made with the reagent suite contained 87.3 percent of the iron and analyzed 65.6 percent Fe and 1.9 percent SiO2 . Continuous flotation tests were made with a nominal feed rate of 400 lb /hr , but without middling recirculation . Using nearly the same amount and complement of reagents as in the batch tests , three similar continuous tests gave concentrates averaging 63.9 percent Fe and 3.1 percent Sio while recovering about 86 percent of the iron .

"Health hazards in the mining industry may be attributed to (1) abnormal conditions of the air, (2) improper sewage disposal, (3) bad drinking water, (4) bad illumination, and (5) local mechanical irritation.Abnormal conditions of the air may be due either to physical or chemical changes. The physical changes are (1) abnormal temperature, (2) abnormal humidity, (3) air movement, and (4) presence of dusts. Dusts may be either poisonous, as salts of lead, arsenic, zinc, or mercury; or nonpoisonous, as dust from coal, silica or quartz, limestone, gypsum, or silicates. Chemical changes may be due to changes in the normal constituents, as lowered oxygen or increased carbon dioxide; or to the addition of abnormal constituents, as carbon monoxide (white damp), hydrogen sulphide (stink damp), sulphur dioxide, methane (marsh gas, fire damp), hydrogen, and nitrogen.Effects of High Temperatures and Humidities The effects of various temperatures and relative humidities have been studied in mines in many countries. Harrington and myself studied them in the metal mines of this country and, in a report written recently, they have been summarized as follows:A. Remaining at rest in saturated air at 91 1/2° F, for one hour, With no air movement caused -1. An increase in body temperature;2. A moderate increase in pulse rate;3. Profuse sweating;4. Lithium is the lightest of all metals, but as it deteriorates rapidly in air it has no structural applications. Alloys of lithium that will float upon water can be made with other light metals, but these ultralight alloys are not useful in aircraft production or elsewhere at present. Alone or alloyed with alkaline earth meals, lithium acts as reining agent on molten metals, and as a minor constituent with other metals it imparts characteristic properties to the allay. Lithium compounds have somewhat wider application in commerce than either the metal or the alloys. They are used in the glass, ceramic, chemical, and air-conditioning industries and less extensively in the optical, pharmaceutical, electrical, and textile industries. The United States is the leading producer anti consumer of lithium ores and compounds and apparently has the largest known ore reserves. The value of lithium ores and primary chemical byproducts produced is relatively small, yet the lithium chemical industry yields products worth hundreds of thousands of dollars each year, owing partly to the high degree of purity demanded by the trade for most finished products and to the low combining weight of lithium. Fortunately, the low-grade ores can now be concentrated economically, and this fact, together with our extensive reserves, assures adequate supplies to meet any reasonable increase in demand. Lithium is listed as essential by both the American and British war agencies, and its military importance may be greatly enhanced.

This paper describing the operations of the Cresson mine of the Cresson Consolidated Gold Mining & Milling Co. at Cripple Creek, Colorado, is one of a series of papers on mining methods and costs being prepared by the United States Bureau of Mines. The Cresson has recently been the most, important mine in the Cripple Creed district. During 1932 and 1933 an average of 230 tons of new ore (excluding dump material) that averaged about $10 in gold was shipped daily; 150 to 175 men were employed. Stoping was by the shrinkage method exclusively. The ore was screened washed, and hand-sorted at the surface and shipped to the Golden Cycle mill at Colorado Springs for treatment. A considerable proportion of the production was made by lessees working under the so-called "split-check" system. The authors are indebted to the management of the Cresson Consolidated Gold Mining & Milling Co. for permission to publish this paper.

Demand for arsenic trioxide dropped markedly during 1983, primarily as a result of a 27% reduction in cotton planting. As an oversupply of arsenic trioxide developed, prices declined for both domestic and imported material. As part of its mission, the U.S. Bureau of Mines continues to develop technologies that will enable cost-effective compliance with environmental requirements. To this end, investigations to improve the kinetics of chalcopyrite dissolution by simultaneous grinding-leaching in ferric sulfate solution were conducted using the Bureau-designed turbo mill. The development of such a hydrometallurgical process would provide a nonpolluting alternative to conventional smelter technology. The effects of mill rotational speed and volume percent solids on the leaching time and energy required for copper extraction from chalcopyrite were evaluated. The results of this laboratory study have shown that the simultaneous grinding-leaching of chalcopyrite concentrate using the turbomill is technically feasible with essentially 100 pet extraction of copper being possible at 90° C. An optimal energy consumption of 1,153 kW?h/st of copper leached was found in batch testing with the mill operating at 400 rpm with a slurry containing 35 vol pet chalcopyrite. This energy consumption, while higher than other proposed leaching processes (about 600 kW?h/st), is of the same order of magnitude. Further refinement of the energy requirements would require additional testing using a continuous circuit. Leaching times of less than 1 h, for 80 pct copper extraction, are achievable using the turbomill grind-leach process.

The Hornsilver lead-zinc property was examined by the Bureau in 1942 and again in 1944. A project was started on the property in April 1946 and completed in January 1947. Silver was first discovered in this district in 1883, and while complete production records are not available, it is known that a substantial tonnage of high-grade silver ore was shipped before 1887. All of the ore was mined from surface or near-surface workings. When the high-grade oxidized ore was exhausted, the district became inactive and remained so until the 1920's. In 1928 and 1929 some activity was shown in the district, but the Hornsilver mine was not reopened until November 1937. Between 1939 and 1946, the mine produced about 16,000 tons of ore of 531 ounces of gold, 283 ounces of silver, over a million pounds of load, and nerdy 2 million pounds of zinc. The property is now controlled by the Era Mining & Development Co., which has acquired the Last Chance and Ella groups of claims, in addition to the Hornsilver mine.

The most promising candidate for the recovery of alumina from non-bauxitic sources is the leaching of clay by HCL acid followed by crystallization of aluminum chloride hexahydrate (ACH) by hydrogen chloride sparging. Hydrothermal precipitation of boehmite ( ?-A100H) from aluminum chloride solution will simplify the process, will reduce energy consumption and will eliminate corrosion problems involved in calcination of ACH to obtain alumina. This report summarizes the results of Phase I of a program to study the precipitation of boehmite from AlC13 solutions. The amount of boehmite precipitation from dilute A1C13 solutions (5 wt%) is . negligible below 200°C. Above 200°C the precipitation increases with increasing temperature approaching 40% at 300°C. Addition of HCl acid to the dilute A1C13 solution even in small quantities drastically reduces boehmite precipitation. Solution impurities such as Fe, Ca, Mg, or Na substantially promote the precipitation of boehmite. No precipitation is observed from concentrated A1C13 solution (30wt%) representing the leach liquor.

The Bureau of Mines studied pressure-buildup data from a number of California gas wells to determine if the reservoir properties calculated from these data correlated with field-observed reservoir properties. Pressure-buildup curves from 24 wells showed distinct changes in slope, indicating changes in permeability of the reservoir rock. Calculated distances from the wells to indicated permeability changes were in fair agreement with distances deter-mined in the field. Pressure-buildup curves from three wells had more than one distinct change in slope, indicating multiple permeability changes. Thus field data supported the theoretical relations used to calculate reservoir properties from pressure-time relations measured in gas wells.

The Bureau of Mines used a diphenyl ether calorimeter to determine the heat contents of natural and synthetic blast-furnace slags. For solid slags, enthalpy (HT-H298) at 1,448° to 1,680° K ranged from 282.1 to 348.5 cal/g (±3 percent); liquid slag values were 400.1 to 514 cal/g (±3 percent) in the temperature range between 1,626° and 1,890° K. Experimental results for liquid slags were 14.3 to 16.2 percent higher than the enthalpies calculated from thermodynamic tables; values for solid slags ranged from 5.3 percent lower to 3.9 percent higher than calculated values. Experimental results for liquid synthetic slags ranged from 14.9 to 24.3 percent higher than the calculated enthalpies; values for solid synthetic slags were from 1.0 to 3.4 percent higher.

The panic bars (and associated linkages and electrical circuitry) that are defined in this document are a machine operator-actuated control for the emergency deenergization of the tram motors on self-propelled, electric and diesel face equipment; including haulage equipment such as shuttle cars. There are nine basic types of panic bars (and several variations) that are recommended in this document, but all of the panic bars are characteristically of the "fire door" type that is frequently encountered on outside exit doors of many public and some commercial buildings. It is also the intent of the following panic bar designs that, when emergency brakes are added to the face equipment machinery for which they arc required, that they be designed and hooked up so that actuation of the panic bar will result in automatic application of the emergency brakes (immediately following the deenergization of the tram motors). The following panic bar design requirements are a restatement of the proposed requirements defined in the Federal Register, requirements cited in "Guidelines for the Design and Installation of Devices for Deenergization of Self-Propelled Electric Face Equipment/Automatic Emergency Brakes" by G. E . Lester and J. D. Greenhalgh of MESA, and Bendix recommendations that were formulated as a result of discussions with personnel at the various machine manufacturers and underground coal mines that we surveyed, discussions with G. L. Judy (our consultant), etc:

[There have been large increases in the oil production and casing-head gas production of certain wells on the Quinn dome (northern part of the Lyons-Quinn field of Oklahoma) caused by large volumes of gas under high pressure entering Vie productive oil sand, The oil production of sore of the leases in this part of the field temporarily increased over 100 per cent within a few days after gas entered the oil sand among the leases that had the greatest increases in oil production wore those on which no gas wells were drilled but which were located up-elope on the structure in reference to gas wells on adjoining properties. It is conservatively estimated that 1,444,000,000, cubic feet of gas entered the productive oil sand and, up to January 1, 1924, caused an increase of about 950,000 barrels of oil from this sand. The casing-held gas volumes of a number of wells producing between 25,000 and 75,000 cubic feet of gas per day increased to several hundred thousand cubic feet and a few walls increased to nearly two million cubic feet per day after extraneous gas entered the oil sand from which the wells were producing.]

This report describes the procedures used by the Bureau of Mines for obtaining attitudes of joints surveyed with a borescope in inclined boreholes. An algorithm for computing the dips and azimuths of the observed joints is presented. The algorithm is used to solve an example problem. A computer program based on the algorithm is listed. It is hoped that the procedures described here will be useful in treating large quantities of data obtained from comprehensive borehole surveys.

"N-acetyl-beta(B)-D-glucosaminidase is a N-acetyl-beta((B)-D-glucosaminidase enzyme se¬creted by alveolar macrophages in response to phagocytosis of particulate material. Alveolar macrophages participate in the degradation and fibrosis of pulmonary tissue that results in pneumoconiosis. Known quantities of four characterized respirable dusts were bronchoscopically placed into the right caudal lung lobe of macaque monkeys. Bronchoalveolar lavage (BAL) samples were collected from dust-exposed right lung and unexposed left lung of the same individuals at 2-week intervals for 12 weeks after dust instillation. The samples were tested for N-acetyl-B-D-glucosaminidase activity to determine if the enzyme levels could serve as an indicator of pulmonary injury induced by generic coal dusts when compared to known fibrogenic and nuisance dusts. Installation of generic quartz, anthracite, or TiO2 dusts produced significant elevations of enzyme activity and increased numbers of macrophages in the dust-exposed lobes. Elevations in enzymatic activity and macrophage numbers were greatest in response to generic quartz dust. These results suggest that quantitative levels of N-acetyl-B-D-glucosaminidase activity may be a useful indicator of acute and chronic lung injury following exposure to fibrogenic and nonfibrogenic dusts.IntroductionCoal workers pneumoconiosis (CWP) is an occupational pulmonary disease associated with the inhalation of coal mine dust. An important external factor in the development of CWP is the composition of the respirable coal mine dust. A cause-and-effect relationship exists between coal mine dust and CWP, but it has not been shown that exposure to generic coal dusts will cause CWP [12]. Numerous immunological, biochemical, and cellular changes occur in CWP; however, in this study, only N-acetyl-beta(B)-D-glucosaminidase (NAGase), protein, and alveolar macrophages (AM) will be examined [7, 32, 33]."

"INTRODUCTION The wartime demand for iron ore and alloy steel on the west coast revived interest in iron reserves in western United States and Alaska. Magnetite was known to have been the most abundant metallic mineral at some of the old copper mines on Prince of Wales Island. In the summer of 1942, the Bureau of Mines began a program of preliminary investigation followed by core-drilling two magnetite deposits on the island.The Poor Man iron deposit on Kasaan Peninsula, Prince of Wales Island, Southeastern Alaska, was examined in August 1942 by Robert L. Thorne, associate mining engineer of the Bureau of Mines, and John C. Reed and B. N. Goddard, geologists of the Federal Geological Survey in September 1942 the area was mapped geologically and a magnetic survey was made by the Geological Survey. In October and November 1942, a more thorough preliminary examination, which included cleaning out and sampling old adits and trenches, was made by thesenior author, Robert L. Thorne, and Aner W. Erickson, engineers of the Bureau. J. G. Shepherd, a consultant of the Bureau, spent two weeks in March and April 1943 investigating the Poor Man and other deposits on Kasaan Peninsula. Core drilling was completed in June 1943.L. A. Warner, of the Geological Survey, examined all cores and interpreted structural data revealed by the drilling."

As part of a general preliminary investigation of lead-zinc deposits in Stevens County, Wash., the Electric Point and Gladstone mines on Gladstone Mountain, in the Northport Mining District, were examined by R. M. Gammell, an engineer of the Bureau of Mines, in June 1946. A trenching and sampling program was executed later the same year under' the supervision of Miro Mihelich and the author. This report presents the factual data obtained from the trenching and sampling operations.

The Bureau of Mines investigated methods of refining iron-contaminated zinc to marketable quality and developed a laboratory-scale process in which aluminum, added as a refining agent, reacted with the iron, forming solid Fe2Al which floated to the surface of the melt. Solids were separated by both filtration and centrifugation, using a unique type of "dipping" centrifuge devised during the work. Typical zinc recovery was 95 percent, and iron content of the metal was lowered from 0.37 to 0.02 percent.
