Physiol. 256 (Cell Physiol. 25): C405-C412, 1989.-A kinetic model was developed to describe the production of superoxide (02) by single pulmonary alveolar macrophages (PAM). Model predictions were compared with experimental results obtained from single rat PAM. The 02 was quantified by measuring the reduction of nitro blue tetrazolium (NBT) to a diformazan precipitate (NBTH,) from video-recorded images of individual cells. The kinetic model considered three reactions: 1 ) the production of extracellular 02 from the reduction of oxygen by NADPH oxidase using intracellular NADPH as the substrate, 2) the subsequent dismutation of 02 to form H,O,, and 3) the reaction of 02 and NBT to form diformazan. NBT specificity for 02 was analyzed by comparing results in the presence and absence of superoxide dismutase (SOD) that catalyzes the dismutation of 02 to H,O,. Measured PAM heterogeneity was accounted for by varying the concentration of intracellular NADPH, its rate of depletion, and the concentration of intracellular NADPH oxidase in the kinetic model. Model predictions compared favorably with experimental results except when SOD was present. This discrepancy may be due to diffusional limitations because NBT is a relatively small molecule (818 mo! wt) compared with SOD (34,000 mol wt). In addition, the cell surface is both ruffled and negatively charged, which may introduce steric hindrances and/ or electrostatic effects, since SOD is also negatively charged, whereas NBT is positively charged.PULMONARY ALVEOLAR MACROPHAGES (PAM) protect the lungs by phagocytizing foreign debris and bacteria. This process is accompanied by the production of superoxide anion (02) at the surface of the plasma membrane (3, 31). Superoxide readily dismutates to form H20 2 and subsequently, other oxygen metabolites which have been shown to be both antibacterial and cytotoxic (21, 25, 35).Superoxide is produced from the reduction of extracellular O, at the surface of the cell membrane by NADPH oxidase by the following reaction (3, 4)"

The linear cutting system, developed by the U.S. Bureau of Mines, uses geometric principles developed by Cardan to produce a nearly constant cut depth. The new system has been extensively tested in a synthetic material under laboratory conditions to verify mechanical capability and to identify operational characteristics. Comparisons between 15-rpm linear cutting and 50-rpm rotary cutting systems show significant improvement in respirable dust entrainment, product size distribution, and energy usage. Respirable dust is reduced by as much as 90 pct. Recovered product showed a 67-pct reduction in -0.32-cm (-1/8-in) material and a 200-pct increase in +5.08 cm (+2 in) material. Average power was reduced by 66 pct for the linear cutting. Because the bit cutting paths differ between linear and rotary cutting, it was necessary to compare the two at the same cut depths and bit types. These comparisons show that low revolution per minute rotary cutting entrains about the same amount of respirable dust as the linear cutting system, but the average shaft torque may be 55 to 130 pct greater for the rotary system.

The Bureau of Mines used fiber-laden air to test two identical fibrous aerosol monitors (FAM) in order to establish the FAM-optical count correlation. The FAM is a unique instrument developed by GCA Corp. (GCA) to measure real-time fibrous aerosol concentrations. It will also respond to particulates that have sufficient shape irregularities to possess fiber characteristics such as the 3:1 length: diameter (L:D) aspect ratio. Results of the Bureau's laboratory tests showed that the FAM response is linearly correlated to concentration data obtained using the optical membrane filter count technique approved by the National Institute for Occupational Safety and Health (NIOSH), U.S. Department of Health, Education, and Welfare. A plot of the FAM-optical count correlation showed that FAM concentration readings are generally well within ±25 pct of the optical membrane filter count. Initial tests to estimate the best ratio control setting of the FAM for 5-µm fiber-length discrimination were performed using one instrument only (the ratio control setting determines the minimum threshold fiber length that will be detected by the FAM). The estimated setting was then used to correlate both FAM units. The result was a 65 pct difference in correlation between the two units. It appears that the major factor responsible for this difference was the internal adjustment of the photomultiplier detector sensitivity for each of the two FAM units.

This paper describes mining operations at the Iron King mine of the Shattuck -Denn Mining Corp. in Yavapai County , Ariz . It is one of a series being prepared by the Mining Division of the Bureau of Mines on mining practices and methods in various mining districts of the United States . The mine and surface plant of the Iron King mine are situated in secs . 15 and 21 , T. 13 N. , R. 1 E. , about 20 miles by State highway 69 east by southeast from the city of Prescott and 1 mile southwest of the mining town of Humboldt on the Prescott -Mayer branch of the Atchison , Topeka & Santa Fe Railroad (fig . 1 ) . A l-mile truck haul brings the mill concentrates (lead and zinc ) to bins on a siding of this line at Humboldt ; the silver -gold bullion is handled separately . Some of the oldest residents in the vicinity of Prescott claim that the original location for the Iron King mine was made in 1880. The earliest recorded production was in 1906-1907 . During the first World War the mine produced fluxing ore for the Humboldt smelter . In 1936 the property was purchased under tax sale , and a mill (140-ton capacity ) was erected in 1938. The capacity of this mill was increased to 225 tons the following year , and a cyanide plant was added in 1940 to treat mill tailings . In 1942 the Shattuck -Denn Mining Corp. purchased the property . A new working shaft was sunk , mine development was speeded , and mill capacity was again increased . The predominating locally formation of this district is a pre -Cambrian schist known as Yavapai schist . The Iron King deposit , which carries gold, silver , lead , zinc , and copper , occurs as a series of lenses in a sheared zone of the pre -Cambrian schist . Down to the 600 -foot level the ore was mined and drawn by the shrinkage method . The cut -and -fill method was adopted for the lower levels . Waste rock for stope fill was provided by block-caving an area on the 600 level . Main haulage drifts were driven in the firm footwall , and crosscuts were driven across the mineralized zone at 100 -foot intervals . Grab samples taken from each car of every working place and channel samples cut every 5 feet in development work are considered sufficient for estimating ore reserves .

To identify construction materials for emerging technology in chemical and metallurgical processes, the Bureau of Mines investigated the acid resistance of ceramic materials. Eight commercial ceramic materials comprising two red shale, two fire clay, a silica, a five-claybonded silicon carbide, a carbon, and a high-alumina brick were evaluated. Samples were exposed for 110 days to 20 wt pct HCl at 50°, 70°, and 90° C and to 30 wt pct HCl at 70° C. Statistically significant changes in cold crushing strength, volume, and weight were generally small or not detected. The leach rates of Al, Ca, Fe, K, Mg, Na, Si, and Ti ions were monitored. The carbon brick sample had the best acid resistant properties with a maximum total leached ion weight loss of 0.004 wt pct. The silicon carbide, the silica, and one of the fire clay bricks also had excellent acid-resistant properties.

Air pollution control regulation$ already implemented or being considered for implementation require substantial investment of capital to remove the sulfur oxides from copper, lead, and zinc smelter gases. There will be an attendant increase in production costs and possibly an associated increase in the price of these metals to the consumer. The total investment and any resultant metal price increase will depend upon the control regulations that ultimately will be adopted by the individual States. Controversy has arisen between the metals industry and governmental control agencies regarding the status of stack gas desulfurization technology and the ability of industry to control sulfur oxide emissions to the degree proposed by some of the more stringent regulations currently under consideration. This paper evaluates the problems involved in achieving various degrees of control over smelter sulfur oxide emissions and the technology available to accomplish the task of assuring clean air. The estimated cost of compliance is reviewed for the copper, zinc, and lead industries, but principal at tent ion is given to the copper situation.

This report describes work done by the Bureau of Mines to simplify the design and operation of the vortex cone, a new device for prerefining blast furnace iron. Three new apparatus arrangements were tested. Results with one of these arrangements average 74.2 pct silicon removal and 61 pet oxygen efficiency (disregarding removal of carbon and manganese).

The Bureau of Mines through its contract research program recently analyzed the mining and environmental requirements in the premining stage for copper mining in the northern highland of Wisconsin. The Critical-Path-Method (CPM) was used in this research. However, a step-by-step derivation of the CPM Diagram was beyond the scope of this effort. The Bureau of Mines objective now is to show, step by step, how the original hand-computed CPM Diagram was constructed. The streamlining (restructuring) of this diagram from the original effort is also summarized. The critical path time (longest time) through the original CPM Diagram was 25.12 yr, excluding public liaison effort. This diagram showed the involvement of the regulatory agency, mining applicant and public in the premining stage. Analysis of this diagram produced a streamlined version which reduced the critical path time by nearly 7 yr. The CPM construction methodology offered here may be applied to mine planning for any mining operation.

The Bureau of Mines developed hypereutectic aluminum-silicon alloys which 11.7.d better mechanical properties than the unmodified hypereutectic alloy. A commercial aluminum-20 weight-percent silicon LIr alloy was selected as a ary base for improvement. The primary silicon. grains were reduced from ASTM 4-5 to ASTM 8.5 by the use of phosphorus and phosphorus compounds. The remaining eutectic matrix was refined by sodium metal additions of 0.2 weight-percent. Solid solution strengthening of the alpha aluminum was obtained by adding 2 percent copper and 0.5 percent magnesium. Also, 1 percent nickel was added to harden the alloy. Optimum thermal treatment consisted of a solution treatment for 10 hour:, at 480° C with hot water quench, followed by aging for 6 hours at 175° C. The final alloy exhibited a Rockwell B hardness of 75 ±2 in the heat-treated condition. The ultimate tensile strength was 39,5O0 psi with no recordable elongation. The coefficient of linear thermal expansion was 20.4 x 10-6 per °C in the temperature range 20° to 300° C. No wear resistance data were collected.

World resources of cobalt appear ample for the foreseeable future; however, political and economic events have caused concern over the availability and reliability of an uninterrupted supply. Because of this concern, the U.S. Congress requested that the U.S. Bureau of Mines initiate a study of the flow of cobalt in the economy that would delineate and quantify the production areas where metal values are lost. As a result, the Bureau has developed a computerized commodity flow model. A major attribute of this model is that it is amenable to updating as supply, demand, and/or production data change. This report follows the flow of cobalt through its metallurgical and chemical applications and highlights areas where significant losses occur due to downgrading, export, or disposal. The study indicates that about 4,300,000 lb of cobalt-bearing materials are lost each year, representing 24 pct of U.S. domestic demand.

Hydrogen sulphide associated with natural gas was noted as early as 1892, at Point Albino, Canada.4 The presence of this impurity was recognized also in other of the earlier oil and gas fields, although the occurrence urea by no means common. However, in recent years, with the development of the producing zones of west Texas and the Texas Panhandle, the production of "sour" crude has increased greatly and hydrogen sulphide has proved a generally troublesome factor--frequently a major problem--in the drilling and operation of many wells in those areas. The problems arising from the drilling and producing of oil or gas wells in hydrogen-sulphide areas, fall under two general divisions: safety problems and corrosion problems. Mention is mane of a third division which is closely related to the other two, that of treating--or the development of processes for the removal of hydrogen sulphide. Field reconnaissance and laboratory tests on clay from South Windsor, Hartford County, Conn., indicate that the clay is suitable for lightweight aggregate raw material. Exploration of the deposit supports the conclusion that the reserves of clay are sufficient for economical mining operations to produce lightweight aggregate. Preliminary tests indicated that the clay has a short bloating range. Initial expansion occurred at 2,000° F, and at 2,100° F the clay was slightly overfired and sticky. Rotary kiln tests verified the preliminary data, and these tests showed that the clay would make a good lightweight aggregate if the processing temperature and the kiln feed sizes were closely controlled. Lightweight concrete made from the processed aggregate had properties comparable to those of concrete made from a commercial clay aggregate.

This paper summarizes precent knowledge of the ignition qualities of pure hydrocarbons likely to be present in modern Diesel engine fuels, as shown by an extensive search of the technical literature. The report also discusses the possible significance of cetane number and hydrocarbon struc- ture. Cetane or cetene numbers, and other physical properties, are tabu- lated for about 100 hydrocarbons, and a bibliography is appended. In recent years the Diesel engine has enjoyed increasing popularity due to its dependability, inherent characteristics of fuel economy, and the fact that it operates on a fuel that hitherto has been available in comparatively large quantities at a low price. These qualities have en- abled the compression-ignition engine to compete very favorably with other types of prime movers in many fields and have led to predictions of fur- ther rapid expansion in the application of Diesel power. In a survey of the field of all petroleum fuels for combustion engines, Holaday et al (9) have predicted that by 1948 railroads will be using 10 times as much Diesel fuel as was used in 1939 and that over-all consumption will have doubled that of 1939. Other estimates of the yearly demand for Diesel fuels five years hence vary from 2,6 to 5 times the amount used in 1944. Into this picture has come the question of whether the greatly in- creased production to meet this estimated demand is compatible with the maintenance and possible improvement in quality of fuels necessary to satisfy future engines. Unfortunately perhaps for the Diesel engine, factors other than those of simple arithmetical multiplication must be considered. For example, the consumption of oil in central heating plants doubled from 1935 to 1943, and a further increase of 57 percent of the 1943 demand is expected by 1950. Jet propulsion has been developed to the stage where it is entirely practical for military aircraft. Catalytic cracking plants, many of which were constructed to meet the war- time accelerated demand for high-octane gasoline, are taking their place in the refining industry. All these developments, as well as the Diesel engine, require material of approximately the same boiling range. Virgin gas oils make excellent burner fuels, perform most satisfactorily in the gas-turbine units for aircraft according to present information, and are at a premium as catalytic cracking stock.

An electronic color-sorting machine was tested for recovering colorless glass concentrates from mixed glass products. The tests were made using glass recovered from unincinerated (raw) urban refuse and residues resulting from the incineration of refuse. The results of the study revealed that concentrates analyzing about 86 percent colorless glass could be recovered from mixed unincinerated glass containing 54 percent colorless glass material. Recovery of the colorless glass was about 62 percent. Color sorting the incinerated glass was somewhat more difficult. Multiple-stage processing was required to recover 59 percent of the colorless glass material in a product analyzing 89 percent colorless glass. The feed material analyzed 41 percent colorless glass.

For nearly 30 years the Bureau of Mines has contended that mine explosions can be prevented and has pointed out to the industry ways and means by which they could be avoided. The record of co2l-mine explosions • during the past fiscal ?Bar bears out the :Bureau1s contention, inasmuch as fewer men were killed by coal-mi~e e:xplosions in the United States during that year than in :gi_y corresponding 12 months in the history of coal mining in the United States, insofar as records are available; and more than a year has elapsed since·a major explosion (in which five or more men were killed) has occurred, this being t.1-J.e first continuous 12- month period without a major coal-mine explosion disaster in the United States in more than 60 years. The methods of preventing mine e:iiplosions are well known to experienced, well-informed persons cngnged in coal mining. Despite this knowledge, many coal-mine operutors continue to use practices and methods that they should know have resulted in countless explosions, and mine workers persist in following past procedure or in breaking safety rules and practices almost certain eventually to result in disaster. ' Virtually all explosions are the result of human failure, such as carelessness or neglect by officials or workers or others. These human failures can be corrected chiefly by ed.ucation and proper and adequate supervision and strict discipline. In some instances mine officials are ·lax, careless, poorl~• informed, and often incompetent; end in many instances miners have repulsed the introduction of known preventive methods because of sheer obstinacy, ignorance, heedlessness, or possibly fear that the operator was trying to o'bta.in some unfair adva.,.'1. tago or would neglect some other phase of accident or e,:plosion prevention. The present work is part of an ongoing Bureau of Mines effort to provide a better understanding of the fundamental phenomena involved in sulfide flotation, the role of oxygen in xanthate adsorption being one essential issue. Galena was chosen as the material around which to develop a conceptual model, partly because of its demonstrated oxygen sensitivity and partly because information on its properties and flotation behavior constitutes the largest part of the fundamental information available on sulfides of commercial importance. The model is formulated primarily in terms of surface electronic energy levels produced by oxygen and xanthate and is further developed using existing experimental evidence and its implications. Four major functions of oxygen are elucidated, and the relationship of these to the formation of lead xanthate, dixanthogen, and a surface compound is discussed. Though yet incomplete, the model provides a useful framework within which the results of diverse experiments can be related to one another in their proper perspective, as shown by a brief discussion and resolution of some apparently discordant results reported in the literature.

The electron optics, vacuum system, and sample stage assembly for the Bureau of Mines? electron probe X-ray spectrograph were purchased. Four scanning curved-crystal spectrometers, two reflection- and two transmission-type, were designed and constructed by the authors. The reflection spectrometers are placed in a vacuum chamber for measurement of long-wave-length X-radiation. Detection, pulse-amplitude discrimination, and recording circuits are provided for the simultaneous measurement of four X-ray spectral lines. Minimum spot size is approximately 2 microns in diameter at 30 kilovolts and a sample-to-ground current of 5 x 10-8 ampere. At this voltage and current the limit of detection with reflection spectrometers is less than 0.05 weight-percent for most elements. The limit of detection for elements determined by transmission-type spectrometers, evaluated at 35 kilovolts and 5 x 10-8 ampere, is approximately 0.1 weight--percent for all elements having excitation voltages below 20 kilovolts. In general, reflection spectrometers exhibited satisfactory but poorer resolution than the transmission type, resolution of the spectrometers being limited primarily by crystal imperfections, with greater mosaic structure being introduced by the grinding process for the reflection crystals. Several applications of the electron probe X-ray spectrograph are described. Analysis of a uranium-steel is discussed in detail to illustrate calculation of composition from measured X-ray intensities.

The discovery of the Oklahoma City field and its .rapid development into one of the major oil-producing districts is an outstanding epoch in the history of the petroleum industry not only of .Oklahoma but also of the United States. The relatively great depth to which the wells were drilled and the large initial production of high-gravity oil are of special significance. The field has boon under proration almost from the time that its development began, and many new production problems have developed out of the changed conditions of operation. The discovery well was first tested in December 1928 at a depth of 6,402 feet, with an initial production of 4,847 barrels of 38.5° A.P.I. gravity and several months later was completed at a depth of 6,624 feet. The large initial production, the high pressure, and the large gas volume in the different horizons of the discovery well suggested the potential possibilities of gas and oil, production in formations of Pennsylvanian and pre-Pennsylvanian age.
